目的:研究外源性 Apelin-13对急性心肌梗死大鼠心肌细胞保护作用。方法:通过结扎左冠状动脉前降支方法建立急性心肌梗死大鼠模型,存活大鼠分为心梗组(AMI 组)及 Apelin-13治疗组,另设假手术组(Sham 组)及假手术 Apelin-13治疗组(Sham+Apelin-13组)。治疗14d 后,取心脏组织,TTC/Evans Blue 染色检测心肌梗死面积;HE 染色检测心脏组织病理变化;TUNEL法检测心肌组织细胞凋亡阳性率;Western blot 法检测凋亡蛋白 Bcl-2、Bax、Cleaved-Caspase-3的表达。结果:与急性心肌梗死模型组相比,Apelin-13治疗后,大鼠心脏组织梗死率显著减小,心肌组织炎性细胞富集减少,凋亡心肌细胞数减少,促凋亡蛋白 Bax、cleaved-Caspase-3表达减少,抗凋亡蛋白 Bcl-2表达增多。结论:Apelin-13可以有效缓解缺血引起的急性心肌梗死和心肌细胞凋亡,可以作为临床治疗急性心肌梗死的备选药物。

목적:연구외원성 Apelin-13대급성심기경사대서심기세포보호작용。방법:통과결찰좌관상동맥전강지방법건립급성심기경사대서모형,존활대서분위심경조(AMI 조)급 Apelin-13치료조,령설가수술조(Sham 조)급가수술 Apelin-13치료조(Sham+Apelin-13조)。치료14d 후,취심장조직,TTC/Evans Blue 염색검측심기경사면적;HE 염색검측심장조직병리변화;TUNEL법검측심기조직세포조망양성솔;Western blot 법검측조망단백 Bcl-2、Bax、Cleaved-Caspase-3적표체。결과:여급성심기경사모형조상비,Apelin-13치료후,대서심장조직경사솔현저감소,심기조직염성세포부집감소,조망심기세포수감소,촉조망단백 Bax、cleaved-Caspase-3표체감소,항조망단백 Bcl-2표체증다。결론:Apelin-13가이유효완해결혈인기적급성심기경사화심기세포조망,가이작위림상치료급성심기경사적비선약물。
Objective:To study the protection effect of Apelin-13 on myocardial cell of acute myocardial infarction in rats.Methods:The acute myocardial infarction methods (AMI)model was developed by ligation of the left anterior descending coronary artery.The surviving rats were divided into AMI group (AMI)and Apelin-13 trea-ted group (Apelin-13),and the groups of sham operation and Apelin-13 treated sham operation were also set up(n=8).After treatment for 14d,the rat hearts were taken,then detected the myocardial infarction area by TTC&Evans Blue staining and the pathological changes of cardiac tissue through HE staining.Furthermore,TUNEL method was used to detect the apoptotic myocardial cells,and the expression of the apoptotic protein Bcl-2,Bax and cleaved-Caspase-3 were detected by Western blotting.Results:Compared with the AMI group,after Apelin-13 treatment, the relative area of infarction myocardium in rats were significantly reduced,the accumulation of inflammatory cell in myocardial tissue and the number of apoptotic myocardial cells were decreased,the expression of pro-apoptotic pro-tein Bax and cleaved-Caspase-3 were increased and the expression of anti-apoptotic protein Bcl-2 was decreased.Con-clusion:Apelin-13 could effectively alleviate the acute myocardial infarction and myocardial cell apoptosis induced by myocardial ischemia,and Apelin-13 can be used as a drug for clinical treatment of acute myocardial infarction.
