为研究番鸭细小病毒基因遗传变异特征,通过 PCR技术获得了番鸭细小病毒结构蛋白基因VP3序列 AH-MDPV-VP3,并将该序列与 GenBank 数据库中登录的9条番鸭、鹅和鸭细小病毒基因相应序列进行了比对分析。测序结果可见番鸭细小病毒 VP3基因全长1605 bp,编码534个氨基酸。氨基酸序列同源性和进化分析表明 AH-MDPV-VP3与国内两个番鸭细小病毒基因 KC171936和 KM093740同源性较高,分别为100%和98.3%,且三者之间亲缘关系较近,来源于共同祖先;而与鹅细小病毒,尤其与台湾分离的鸭细小病毒基因 AY382891和 AY382892同源性相对较低,分别为91.6%和90.8%,遗传距离也相对较远。同时可见番鸭、鹅和鸭细小病毒的 VP3基因各自较为保守,仅有少数氨基酸发生变异。本研究初步获得番鸭细小病毒 VP3基因分子特征,为下一步该基因在免疫诊断及免疫防治中的应用奠定了基础。

To investigate variation status of muscovy duck parvovirus gene,the VP3 gene was cloned by PCR,which we named as AH-MDPV-VP3.And we compared AH-MDPV-VP3 with the sequence of nine VP3 genes of muscovy duck,goose and duck parvovirus in the GenBank database.By alignment of the ten gene sequences,the higher homologies between AH-MDPV-VP3 and the KC171936 and KM093740 se-quences of muscovy duck parvovirus isolated in China were found,100% and 98.3% repectively,indcating a close genetic relationship with a common ancestor of evolution.And the homologies with goose parvovir-us were lower,particularly,the AY382891 and AY382892 sequences of duck parvovirus isolated in Taiwan were 91.6% and 90.8%,the genetic distances also were farther.Moreover the VP3 gene of muscovy duck,goose and duck parvovirus were relatively conservative with several amino acid mutations.The mo-lecular characteristics of muscovy duck parvovirus VP3 gene were preliminarily obtained.And this study laid the foundation of immune diagnosis and prevention and control of muscovy duck parvovirus.
