目的::观察参附芎泽方对冠脉结扎所致心衰大鼠血流动力学及心肌纤维化的干预作用并探讨其相关机制。方法:SD大鼠随机分为3组:假手术组、模型组、方药组。模型组、方药组结扎冠状动脉左前降支,假手术组只穿线不结扎。方药组每次经口灌服参附芎泽浓缩液;假手术组、模型组灌服等剂量生理盐水。各组干预8周后采用血流动力学检测评价心功能, Masson染色观察心肌纤维化程度,Western blotting 检测心肌 TGF-β1表达的变化。结果:与模型组相比,方药组左室舒张末压(LVEDP)显著降低,左室收缩压(LVSP)、左室内压上升和下降的最大速率(±dp/dtmax)明显升高;同时,方药组可使心衰大鼠心肌胶原纤维的沉积减少,心肌TGF-β1的表达降低。结论:参附芎泽方可改善冠脉结扎所致心衰大鼠的心功能,抑制心肌纤维化,其机制可能与降低心肌TGF-β1表达有关。

목적::관찰삼부궁택방대관맥결찰소치심쇠대서혈류동역학급심기섬유화적간예작용병탐토기상관궤제。방법:SD대서수궤분위3조:가수술조、모형조、방약조。모형조、방약조결찰관상동맥좌전강지,가수술조지천선불결찰。방약조매차경구관복삼부궁택농축액;가수술조、모형조관복등제량생리염수。각조간예8주후채용혈류동역학검측평개심공능, Masson염색관찰심기섬유화정도,Western blotting 검측심기 TGF-β1표체적변화。결과:여모형조상비,방약조좌실서장말압(LVEDP)현저강저,좌실수축압(LVSP)、좌실내압상승화하강적최대속솔(±dp/dtmax)명현승고;동시,방약조가사심쇠대서심기효원섬유적침적감소,심기TGF-β1적표체강저。결론:삼부궁택방가개선관맥결찰소치심쇠대서적심공능,억제심기섬유화,기궤제가능여강저심기TGF-β1표체유관。
Obiective:To explore therapeutic effects of the Shenfu Xiongze Decoction on hemodynamics and myocardiac fibrosis in rats with heart failure (HF)induced coronary artery ligature.Methods:SD rats were randomly di-vided into three groups:sham operated group,model group and herbs group.The rats of modle group and herbs group received the ligation of left anterior descending artery,while the rats of sham group were accepted sham operation (with-out ligation of left anterior descending artery,the rest of the operation was the same as the model).The herbs group were fed with Shenfu Xiongze Decoction,sham group and model group were fed with saline.After 8 weeks,hemodynamic e-valuation were conducted to determine the cardiac functions.Masson staining was used to observe the degree of myocardial fibrosis.The protein level of transforming growth factors-β1 (TGF-β1 )was detected by Western blotting.Results:Com-pared with model group,left ventricular end-diastolic pressure (LVEDP)decreased in the herbs group.At the same time,left ventricular systolic pressure (LVSP)and left ventricular pressure maximum rising and falling rates (±dp/dt-max)increased significantly in the herbs group.Herbs reduced the degree of myocardial collagen deposition (P <0.01) and decreased the expression of TGF-β1 .Conclusion:Shenfu Xiongze Decoction improves heart functions and inhibits myocardial fibrosis in rats with HF induced by coronary artery ligature by decreasing TGF-β1 protein expression level.
