目的:对豫医卷毛大鼠的Krt71基因进行比较生物学分析。方法:采用PCR方法对豫医卷毛大鼠Krt71的cDNA序列进行克隆、测序,并与BN大鼠、小家鼠、家牛、家绵羊、家猫、苏门答腊猩猩和人的Krt71 cDNA序列及其编码产物的氨基酸序列进行同源性比较和系统遗传学分析。结果:获得了豫医卷毛大鼠Krt71基因全长1573 bp的cDNA序列( GenBank登录号:KF303588)。豫医卷毛大鼠与BN大鼠、小家鼠、家牛、家绵羊、家猫、苏门答腊猩猩和人的同源性分别为99.9%、95.2%、88.5%、87.5%、88.0%、88.1%、88.1%,氨基酸序列的同源性分别为99.9%、98.9%、92.4%、91.2%、91.4%、92.3%、92.1%。结论:Krt71基因在哺乳动物进化过程中具有较高的保守性,是一个具有重要功能的“持家基因”。
目的:對豫醫捲毛大鼠的Krt71基因進行比較生物學分析。方法:採用PCR方法對豫醫捲毛大鼠Krt71的cDNA序列進行剋隆、測序,併與BN大鼠、小傢鼠、傢牛、傢綿羊、傢貓、囌門答臘猩猩和人的Krt71 cDNA序列及其編碼產物的氨基痠序列進行同源性比較和繫統遺傳學分析。結果:穫得瞭豫醫捲毛大鼠Krt71基因全長1573 bp的cDNA序列( GenBank登錄號:KF303588)。豫醫捲毛大鼠與BN大鼠、小傢鼠、傢牛、傢綿羊、傢貓、囌門答臘猩猩和人的同源性分彆為99.9%、95.2%、88.5%、87.5%、88.0%、88.1%、88.1%,氨基痠序列的同源性分彆為99.9%、98.9%、92.4%、91.2%、91.4%、92.3%、92.1%。結論:Krt71基因在哺乳動物進化過程中具有較高的保守性,是一箇具有重要功能的“持傢基因”。
목적:대예의권모대서적Krt71기인진행비교생물학분석。방법:채용PCR방법대예의권모대서Krt71적cDNA서렬진행극륭、측서,병여BN대서、소가서、가우、가면양、가묘、소문답석성성화인적Krt71 cDNA서렬급기편마산물적안기산서렬진행동원성비교화계통유전학분석。결과:획득료예의권모대서Krt71기인전장1573 bp적cDNA서렬( GenBank등록호:KF303588)。예의권모대서여BN대서、소가서、가우、가면양、가묘、소문답석성성화인적동원성분별위99.9%、95.2%、88.5%、87.5%、88.0%、88.1%、88.1%,안기산서렬적동원성분별위99.9%、98.9%、92.4%、91.2%、91.4%、92.3%、92.1%。결론:Krt71기인재포유동물진화과정중구유교고적보수성,시일개구유중요공능적“지가기인”。
Aim:To study the difference of Keratin 71(Krt71) gene and its coding between Yuyi curly hair rats and other species .Methods:PCR technique was developed to clone the sequence of cDNA of Krt 71 gene from Yuyi curly hair rats, and biologic characters of nucleotide and amino acid sequence encoding Krt 71 gene in different species ( rattus norve-gicus, mus musculus, bos taurus, ovis aries, felis catus, pongo abelii and homo sapiens ) were analyzed.Results:A full-length cDNA of Yuyi curly hair rats was 1 573 bp( Accession Number:KF303588 ) .The Krt71 gene nucleotide homology of Yuyi curly hair rats to rattus norvegicus , mus musculus, bos taurus, ovis aries, felis catus, pongo abelii,and homo sapiens was 99.9%,95.2%,88.5%,87.5%,88.0%,88.1% and 88.1%,and those of amino acid sequences was 99.9%, 98.9%,92.4%,91.2%,91.4%,92.3%and 92.1%,respectively.Conclusion:A high degree of conservation of Krt 71 gene exists among different mammalian taxons , and Krt71 gene is an important housekeeping gene .
