目的:探讨睾酮受体( AR)蛋白表达水平的改变对高血压大鼠血管重构( VR)的影响及其信号转导机制。方法:将30只大鼠随机分为对照组、假手术组、高血压组3组(每组10只)。采用“两肾一夹”法制备肾血管性高血压大鼠模型。 HE染色观察主动脉组织形态结构,免疫组化法检测3组大鼠主动脉组织中AR、丝裂原活化蛋白激酶磷酸酶-1(MKP-1)蛋白表达水平。结果:与对照组相比,假手术组主动脉形态结构基本正常,AR、MKP-1蛋白表达水平无显著改变(P>0.05);与对照组及假手术组相比,AR和MKP-1蛋白表达水平下降(P<0.001);且在高血压组AR与MKP-1蛋白的表达呈正相关(r=0.657,P=0.039),在对照组及假手术组无关(r=0.589和0.436, P>0.05)。结论:高血压VR过程中存在AR及MKP-1蛋白表达水平的改变。

목적:탐토고동수체( AR)단백표체수평적개변대고혈압대서혈관중구( VR)적영향급기신호전도궤제。방법:장30지대서수궤분위대조조、가수술조、고혈압조3조(매조10지)。채용“량신일협”법제비신혈관성고혈압대서모형。 HE염색관찰주동맥조직형태결구,면역조화법검측3조대서주동맥조직중AR、사렬원활화단백격매린산매-1(MKP-1)단백표체수평。결과:여대조조상비,가수술조주동맥형태결구기본정상,AR、MKP-1단백표체수평무현저개변(P>0.05);여대조조급가수술조상비,AR화MKP-1단백표체수평하강(P<0.001);차재고혈압조AR여MKP-1단백적표체정정상관(r=0.657,P=0.039),재대조조급가수술조무관(r=0.589화0.436, P>0.05)。결론:고혈압VR과정중존재AR급MKP-1단백표체수평적개변。
Aim:To explore the effect of the change of androgen receptor ( AR) protein expression level on hypertensive vascular remodeling and the underlying mechanisms .Methods:Thirty rats were randomly allocated into 3 groups:control group, sham operation group and hypertensive group (10 rats in each group).The renovascular hypertensive rats model was established using the "2-kidney and 1-clip"method.Using tail-cuff method to measure the systolic blood pressure (pSB) and HE staining to observe the morphological structure of aorta , and the expressions of AR and MKP-1 were detected by immuno-histochemical technique .Results:For the morphological structure , there was no significant difference between the control group and the sham operation group , and also no significant difference in the expressions of AR and MKP-1 was found be-tween the two groups(P>0.05).Compared with the control group and the sham operation group ,the vascular remodeling of the aorta in the hypertensive group was remarkable , and the expressions of AR and MKP-1 significantly decreased ( P<0.001).In the hypertensive group, the expression of AR was positively correlated with the expression of MKP -1(r=0.657, P=0.039), and no such correlation was found in the control group or in the sham operation group (r=0.589,0.436,P>0.05).Conclusion:The vascular remodeling during hypertension is accompanied by the down-regulation of AR and MKP-1.
