目的::观察丹参酮ⅡA 对脑出血大鼠灶周 Nestin、Neun 阳性细胞数影响。方法:78只Sprague Dawley大鼠被随机分为4组,其中正常组6只,假手术组24只,对照组24只,治疗组24只,治疗组腹腔注射丹参酮ⅡA,立体定向仪自体血注入法制备大鼠脑出血模型,免疫组化染色观察正常组及假手术组、对照组、治疗组术后第3d、7d、14d、28d灶周Nestin、Neun的表达及神经功能评分并进行比较。结果:脑出血灶周 Nestin 阳性细胞数从造模后即增多,第7天时明显增多,随后持续升高,并于第14d时数量达到峰值,随后逐渐减少,第28天时仍高于正常水平;治疗组与对照组同期比较,在第7天(P<0.05)及第14天(P<0.01)差异显著。Neun阳性细胞数在造模后3 d 即有明显增多,至第7天时达高峰,此后逐渐下降,至第28天时仍高于正常水平;治疗组与其他组同期比较,有明显差异(P<0.01)。治疗组大鼠神经功能恢复较对照组有明显改善(P<0.05)。结论:丹参酮ⅡA 可诱导脑出血大鼠灶周内源性神经干细胞增殖分化并改善神经功能。

목적::관찰단삼동ⅡA 대뇌출혈대서조주 Nestin、Neun 양성세포수영향。방법:78지Sprague Dawley대서피수궤분위4조,기중정상조6지,가수술조24지,대조조24지,치료조24지,치료조복강주사단삼동ⅡA,입체정향의자체혈주입법제비대서뇌출혈모형,면역조화염색관찰정상조급가수술조、대조조、치료조술후제3d、7d、14d、28d조주Nestin、Neun적표체급신경공능평분병진행비교。결과:뇌출혈조주 Nestin 양성세포수종조모후즉증다,제7천시명현증다,수후지속승고,병우제14d시수량체도봉치,수후축점감소,제28천시잉고우정상수평;치료조여대조조동기비교,재제7천(P<0.05)급제14천(P<0.01)차이현저。Neun양성세포수재조모후3 d 즉유명현증다,지제7천시체고봉,차후축점하강,지제28천시잉고우정상수평;치료조여기타조동기비교,유명현차이(P<0.01)。치료조대서신경공능회복교대조조유명현개선(P<0.05)。결론:단삼동ⅡA 가유도뇌출혈대서조주내원성신경간세포증식분화병개선신경공능。
Obj ective:To observe the effects of Tanshinone ⅡA to Nestin and Neun’s positive cells in perihe-matoma with intracerebral hemorrhage models of rats .Methods:78 Sprague Dawley rats were randomly divided into four groups:normal group 6,sham-operated group 24,control group 24,treatment group 24,treatment group was given an in-traperitoneal inj ection of Tanshinone ⅡA.Stereotaxic apparatus and infuse fresh quantitative autologous blood into rat’s brain was utilized to build up the ICH model.On 3d,7d,14d,and 28d after ICH,the rats were performed behavioral test, and the expression of Nestin and Neun’s positive cells around hematoma in ICH was analyzed by immunohistochemistry. Results:The Nestin’s positive cells started rising at the begining post operation,increased significantly at 7d,the peak ap-peared at 14d,then the number gradually decline,but there were still significant different compared with other groups at 28d,compared with control group,the expression of Nestin’s positive cells of treatment group were more obvious and early,especially at 7d (P<0.05)and 14d (P<0.01).The Neun’s positive cells started rising at the 3d ,the peak appeared at 7d,then dropped gradully,at 28d ,there were still higher compared with other groups.The expression of Neun’s posi-tive cells of treatment group were more obvious(P<0.01).The rat’s neural functional recovery of treatment group was more apparent than the control group(P<0.05).Conclusion:Tanshinone ⅡA can activate endogenous neural stem cell proliferation and improve the neurological function.
