目的 观察替莫唑胺(TMZ)化疗单用或联合Hepatopoietin Cn(HPPCn)对U251细胞增殖活性的影响.方法 通过外源补充HPPCn和诱导HPPCn低表达两种方法观察HPPCn对U251的增敏作用,首先构建慢病毒低表达载体si-HPPCn和它的阴性对照重组质粒scramble,分别转染胶质瘤细胞U251,分别称为低表达组和阴性对照组,两组均设置不同浓度TMZ(0、6.25、12.50、25.00、50.00、100.00 mg/L).外源补充HPPCn实验同时进行,以TMZ化疗单用作为TMZ与HPPCn联合作用的对照组,两种方法均通过细胞计数试剂盒(CCK-8)实验观察细胞增殖抑制.结果 TMZ化疗单用时,U251细胞的吸光度值从1.755 ±0.010逐渐降至0.725 ±0.010,细胞活性随着TMZ浓度的增加逐渐降低;100 mg/L的TMZ作用后,si-HPPCn低表达组的细胞活性(吸光度值为0.843±0.040)显著高于其阴性对照组scramble(吸光度值为0.661±0.030,P<0.05),细胞活性最高提升了1.3倍;外源性补充50 μg/L的HPPCn和不同浓度的TMZ时,其细胞活性(吸光度值为0.224±0.010)显著低于TMZ单用对照组(吸光度值为0.389±0.010,P<0.05),细胞活性最高降低1.7倍.结论 TMZ对胶质瘤细胞U251具有生长抑制作用,且呈浓度依懒性;HPPCn在体外实验中对U251细胞具有化疗增敏作用.

Objective To study the influence of temozolomide (TMZ) chemotherapy alone or in combination with hepatopoietin Cn (HPPCn) on proliferation activity of U251 cells.Methods By exogenous supplementation of HPPCn and induction of HPPCn lower expression, the sensitization effect of HPPCn on U251 cells was observed.Lentivirus expression vector si-HPPCn and its negative control recombinant plasmid scramble were transfected into U251 cells, respectively, serving as low expression group and negative control group respectively.In the two groups we set different concentrations of TMZ (0, 6.25, 12.50, 25.00, 50.00, and 100.00 mg/L).Exogenous supplementation of HPPCn experiment was carried out at the same time.TMZ chemotherapy alone was used as the control group of TMZ in combination with HPPCn.The inhibition rate of proliferation in the two methods was confirmed by cell counting kit-8 (CCK-8) assay.Results The absorbance value of U251 cells gradually decreased from 1.755 ± 0.010 to 0.725 ± 0.010, and the activity of U251 cells decreased gradually with the increase of concentrations of TMZ when TMZ used alone.The activity of U251 cells in si-HPPCn low expression group (the absorbance value was 0.843 ± 0.040) was significantly higher than in negative control group (the absorbance value was 0.661 ±0.030, P <0.05) when TMZ (100 mg/L) was used.The activity of U251 cells when exogenous HPPCn (50 μg/L) was added (the absorbance value was 0.224 ± 0.010) was significantly lower than TMZ alone (the absorbance value was 0.389 ±0.010, P <0.05).Conclusion TMZ can inhibit the growth of U251 cells in a concentration-dependent manner, and HPPCn has the chemosensitization effect on U251 cells in vitro.
