目的 检测人肺腺癌细胞株、组织及其对应的癌旁肺组织中突触小体相关蛋白(SNAP47)的表达,研究SNAP47与肺腺癌发生和侵袭转移的关系,探讨其在肺腺癌发生、发展中的作用.方法 选取常见人肺腺癌细胞株并随机选取有完整临床资料的50例肺腺癌组织和对应的癌旁肺组织,应用实时定量聚合酶链反应(Real-time PCR)法检测SNAP47 mRNA的表达.结果 SNAP47 mRNA在肺腺癌细胞株中的表达明显高于正常肺上皮细胞株(P<0.05);50例肺腺癌组织中33例(66%) SNAP47 mRNA表达水平明显高于对应的癌旁肺组织,差异有统计学意义(P<0.05),且SNAP47 mRNA表达与淋巴结转移、病理分期相关.结论 SNAP47 mRNA高表达可能与肺腺癌的发生有关,参与了肺腺癌的浸润及淋巴转移.

Objective To investigate the expression of synaptosomal-associated protein 47 (SNAP47) mRNA in human lung adenocarcinoma cell lines, lung adenocarcinoma tissues and adjacent normal lung tissues, evaluate the correlation of SNAP47 with the occurrence, invasion and metastasis of lung adenocarcinoma and explore its role in occurrence and progression of lung adenocarcinoma.Methods SNAP47 mRNA expressions in random selected 50 cases of lung adenocarcinomas and their corresponding normal tissues plus 5 lung cancer cell lines were measured by Real-time quantitative polymerase chain reaction (Real-time PCR).Results SNAP47 mRNA expressions were significantly elevated in nonsmall cell lung cancer (NSCLC) tissues when compared with corresponding adjacent normal tissues as well as lung cancer cell lines compared with normal bronchial epithelial cell line.Among the 50 cases of lung adenocarcinoma, 33 (66%) cases SNAP47 mRNA expression levels were significantly higher than the corresponding adjacent lung tissue (P < 0.05).Increased SNAP47 mRNA expressions were associated with lymph node invasion and advanced pathological stage (P < 0.05).Conclusion Our data indicated that elevated SNAP47 mRNA expression may be associated with the occurrence and progress of lung adenocarcinoma.
