目的 探讨9-顺式视黄酸(9-cis RA)抑制人胆管癌细胞株QBC939增殖与迁移的能力及其机制.方法 体外培养QBC939细胞株,采用噻唑蓝(MTT)、TOPflash荧光素酶报告基因法、实时定量聚合酶链反应(Real-time PCR)、细胞划痕、Transwell迁移实验以及Western blot法检测9-cis RA对QBC939存活率、Wnt信号通路和上皮-间充质转化(EMT)相关标志物表达影响,以及抑制Wnt信号通路对QBC939的迁移能力的影响.结果 人胆管癌细胞株QBC939经9-cis RA处理后,其增殖、迁移能力和Wnt信号通路活性减弱,上皮细胞标志物E-cadherin蛋白表达升高;抑制Wnt/β-catenin人胆管癌细胞株QBC939的迁移能力降低.结论 9-cis RA抑制胆管癌细胞增殖和迁移的作用可能与9-cis RA对Wnt/β-catenin信号通路的抑制相关.

Objective To investigate the mechanisms underlying 9-cis retinoic acid (9-cis RA) inhibiting cholangiocarcinoma cell QBC939 proliferation and migration.Methods TOPflash luciferase assays, real-time quantitative polymerase chain reaction (Real-time PCR), Transwell assay, wound strack and Western blotting were used to detect the relative growth rate of QBC 939, the activity of the Wnt-signalling pathway, and the ability of cell migration after Wnt-signalling pathway was inhibited.Results Cholangiocarcinoma cell QBC939 proliferation, migration and the activity of the Wnt-signalling pathway were inhibited after the cells were treated with 9-cis RA.However, the expression of epithelial cell marker was increased.Cell migration was inhibited after the Wnt-signalling pathway was inhibited.Conclusion These results suggest that 9-cis RA inhibited Wnt/β-catenin signal pathway, proliferation and migration in QBC939 cells.
