目的:建立高效液相色谱(HPLC)法测定宜昌胡颓子根中槲皮素的含量.方法:采用反相高效液相法测定宜昌胡颓子根中槲皮素含量的,色谱柱为Agilent Promosil C18(250 mm×4.6 mm,5 μm),洗脱程序为梯度洗脱,流速为1 mL/min,检测波长为238 nm;柱温为30 ℃,进样量为10 μL,观察该方法的适用性、精密度、稳定性及重复性等指标.结果:槲皮素样品在考查浓度范围内线性关系良好(r=0.9999);平均回收率为99.66%.HPLC法测定宜昌胡颓子根中槲皮素含量的适用性、精密度、稳定性及重复性均较好.结论:HPLC法简便、准确、重复性好,可用于宜昌胡颓子根中槲皮素的含量测定.

Objective:To establish a HPLC method for the content determination of quercetin in elaeagnus henryi root.Method:RP-HPLC method was used to measure the content of quercetin in elaeagnus henryi root.The determination was performed on Agilent Promosil C18(250 mm×4.6 mm,5 μm)with gradient elution at the flow rate of 1.0 mL/min.The detection wavelength was 238 nm.The column temperature was set at 30 ℃.The injection volume was 10 μL.The indexes of this method such as applicability,accuracy,stability,repeatability and so on were observed.Result:The linear relation of the quercetin was fairly good(r=0.9999) and the average recovery was 99.66%.The applicability, accuracy,stability and repeatability of the HPLC method were all good.Conclusion:The HPLC method is simple, accurate and reproducible,which is suitable for the content determination of elaeagnus henryi root.
