为了解乌鳢( Channa argus)性别分化相关机制,采用RT-PCR和RACE法克隆了乌鳢脑型芳香化酶基因Cyp19a1b cDNA部分序列,运用半定量RT-PCR和qPCR技术检测其在雌雄个体组织中的表达情况。结果显示,乌鳢Cyp19a1b cDNA部分序列为1432 bp,包括120 bp 3′-UTR和1311 bp编码区,编码436个氨基酸。序列比对表明乌鳢Cyp19a1b氨基酸序列包含三个功能保守区,与毛足鲈、平鲷和金钱鱼Cyp19a1b同源性较高,分别为89%、88%和88%。半定量RT-PCR和qPCR结果显示,乌鳢Cyp19a1b基因在雌、雄成鱼垂体中表达量高,在肠组织中表达量低,在雄鱼脑组织中呈现高表达而在雌鱼脑组织中则未表达,在其他组织均未检测到表达,表明乌鳢Cyp19a1b基因的表达存在组织特异性及功能差异。

In order to explore sex differentiation mechanism of Channa argus, we cloned partial cDNA sequence of Cyp19a1b gene using RT-PCR and RACE methods, and detected its expression patterns in various tissues of adult female and male individuals based on semi-quantitative RT-PCR and qPCR techniques .The results showed that the length of the partial Cyp19a1b cDNA was 1 432 bp, including a 120 bp 3′-UTR and a 1 311 bp coding region that encoded 436 amino acids.Alignment of sequences revealed that Cyp19a1b amino acid sequence of C.argus contained three functionally con-servative regions , and had a high level of homology with Trichogaster trichopterus ( 89%) , Rhabdosargus sarba ( 88%) and Scatophagus argus (88%).Semi-quantitative RT-PCR and qPCR revealed that Cyp19a1b mRNA had high levels of expression in the pituitary and low levels of expression in the intestine of both female and male .There was a high expres-sion level in the brain of male , but no expression in the brain of female and in other detected tissues of both sexes .The tissue distribution of gene expression reveals potential tissue specificity and functional differences for Cyp19a1b in C.argus.
