目的:观察清热消炎颗粒对内毒素致发热大耳白兔的解热作用及对血清中TNF-α、IL-1β、IL-6、前列腺素(PGE2)表达水平的影响,探讨清热消炎颗粒的解热机制。方法:体温合格的大耳白兔36只随机分为6组,分别为对照组,模型组,阿司匹林组,清热消炎颗粒低、中、高剂量组。一次灌胃给予药物(阿司匹林、清热消炎颗粒)后大耳白兔耳缘静脉注射内毒素溶液制备发热模型。监测给药前、内毒素注射后1、2、3、4 h体温。内毒素注射4 h后,测定血清TNF-α、IL-1β、IL-6、PGE2水平。结果:清热消炎颗粒中、高剂量组,在内毒素注射后1、2、3、4 h直肠温度升高值均显著低于模型组(P<0.05或P<0.01)。而低剂量组与模型组比较,直肠温度无显著性差异。内毒素致热后4 h,模型组大耳白兔血清TNF-α、IL-1β、IL-6、PGE2水平较对照组显著升高(P<0.01);与模型组比较,清热消炎颗粒高剂量组TNF-α、IL-1β、IL-6、PGE2水平均显著降低(P<0.05)。结论:中、高剂量清热消炎颗粒有明显解热作用,低剂量药物无显著降温作用。清热消炎颗粒高剂量组可明显降低内毒素致热大耳白兔血液的内生致热原TNF-α、IL-1β、IL-6及PGE2。
目的:觀察清熱消炎顆粒對內毒素緻髮熱大耳白兔的解熱作用及對血清中TNF-α、IL-1β、IL-6、前列腺素(PGE2)錶達水平的影響,探討清熱消炎顆粒的解熱機製。方法:體溫閤格的大耳白兔36隻隨機分為6組,分彆為對照組,模型組,阿司匹林組,清熱消炎顆粒低、中、高劑量組。一次灌胃給予藥物(阿司匹林、清熱消炎顆粒)後大耳白兔耳緣靜脈註射內毒素溶液製備髮熱模型。鑑測給藥前、內毒素註射後1、2、3、4 h體溫。內毒素註射4 h後,測定血清TNF-α、IL-1β、IL-6、PGE2水平。結果:清熱消炎顆粒中、高劑量組,在內毒素註射後1、2、3、4 h直腸溫度升高值均顯著低于模型組(P<0.05或P<0.01)。而低劑量組與模型組比較,直腸溫度無顯著性差異。內毒素緻熱後4 h,模型組大耳白兔血清TNF-α、IL-1β、IL-6、PGE2水平較對照組顯著升高(P<0.01);與模型組比較,清熱消炎顆粒高劑量組TNF-α、IL-1β、IL-6、PGE2水平均顯著降低(P<0.05)。結論:中、高劑量清熱消炎顆粒有明顯解熱作用,低劑量藥物無顯著降溫作用。清熱消炎顆粒高劑量組可明顯降低內毒素緻熱大耳白兔血液的內生緻熱原TNF-α、IL-1β、IL-6及PGE2。
목적:관찰청열소염과립대내독소치발열대이백토적해열작용급대혈청중TNF-α、IL-1β、IL-6、전렬선소(PGE2)표체수평적영향,탐토청열소염과립적해열궤제。방법:체온합격적대이백토36지수궤분위6조,분별위대조조,모형조,아사필림조,청열소염과립저、중、고제량조。일차관위급여약물(아사필림、청열소염과립)후대이백토이연정맥주사내독소용액제비발열모형。감측급약전、내독소주사후1、2、3、4 h체온。내독소주사4 h후,측정혈청TNF-α、IL-1β、IL-6、PGE2수평。결과:청열소염과립중、고제량조,재내독소주사후1、2、3、4 h직장온도승고치균현저저우모형조(P<0.05혹P<0.01)。이저제량조여모형조비교,직장온도무현저성차이。내독소치열후4 h,모형조대이백토혈청TNF-α、IL-1β、IL-6、PGE2수평교대조조현저승고(P<0.01);여모형조비교,청열소염과립고제량조TNF-α、IL-1β、IL-6、PGE2수평균현저강저(P<0.05)。결론:중、고제량청열소염과립유명현해열작용,저제량약물무현저강온작용。청열소염과립고제량조가명현강저내독소치열대이백토혈액적내생치열원TNF-α、IL-1β、IL-6급PGE2。
O bjective:To observe the antipyretic effect of Qingrexiaoyan particles on the pyrexial rabbits induced by endotoxin ,and the influence on expression levels of TNF-α, IL-1β, IL-6, PGE2 and further explore the antipyretic mechanism of Qingrexiaoyan particles. Methods:Thirty six rabbits with normal temperature were randomly divided into six groups:control group, model group, aspirin group, Qingrexiaoyan particles low dose group, middle dose group and high dose group. The fever models were induced by injecting endotoxin into the ear vein of rabbits after a lavage for drugs. Temperature was monitored separately before the administration and after the endotoxin injection 1,2,3,4 h. After endotoxin injection 4 h, the serum TNF-α, IL-1β, IL-6, PGE2 levels were assayed. Results:Rectal temperature of Qingrexiaoyan particles middle dose group and high dose group increased continuously after the endotoxin injection, 1,2,3,4 h after injection, rectal temperature were significantly lower than the model group (P<0.05或P<0.01). No significant difference was found between low-dose group and model group. After endotoxin injection 4 h, the serum TNF-α, IL-1β, IL-6, PGE2 levels of model group were higher than the control group (P<0.01);compared with the model group, the TNF-α, IL-1β, IL-6, PGE2 levels of high Qingrexiaoyanning particles dose group were significantly lower (P<0.05). Conclusion:The results show that Qingrexiaoyan particles middle dose group and high dose group have marked antipyretic effect but the low dose group has no such effect. The high-dose group of Qingrexiaoyan particles can significantly reduce the endogenous pyrogen of endotoxin-induced fever rabbits, such as TNF-α, IL-1β, IL-6 and PGE2.
