目的:通过对哮喘患儿血清中血清25羟基维生素 D 水平及维生素 D 受体基因多态性分析,拟探讨25-(OH)-VD 在哮喘发病中的作用。方法本院门诊随诊的40例哮喘患儿为哮喘组,健康体检的儿童40例为对照组。采用酶联免疫吸附试验检测两组患儿血清25羟基维生素 D 水平,采用 DNA 测序技术检测两组患儿 VDR 基因位点rs2228570、rs7975232、rs11574114、rs1544410、rs731236、rs757343有无多态性存在。结果哮喘组儿童血清25-(OH)-VD 水平均低于正常对照组,差异有统计学意义(t=7.498,P <0.001)。VDR 基因序列 rs7975232(ApaI)基因型 GG、GT,rs731236(TaqI)基因型 TT、TC,rs11574114基因型 GG、GC、CC,rs1544410(Bsm I)基因型 GG、GA, rs757343(Tru9I)基因型 GG、GA、AA,在两组间基因型的分布有明显的差异(P <0.01或 P <0.05)。结论哮喘患儿存在血清25-(OH)-VD 不足或缺乏,VDR 基因多态位点 rs7975232、rs1544410、rs731236、rs11574114、rs757343基因型及等位基因分布在两组间存在差异,其多态性可能与哮喘发病相关。

목적:통과대효천환인혈청중혈청25간기유생소 D 수평급유생소 D 수체기인다태성분석,의탐토25-(OH)-VD 재효천발병중적작용。방법본원문진수진적40례효천환인위효천조,건강체검적인동40례위대조조。채용매련면역흡부시험검측량조환인혈청25간기유생소 D 수평,채용 DNA 측서기술검측량조환인 VDR 기인위점rs2228570、rs7975232、rs11574114、rs1544410、rs731236、rs757343유무다태성존재。결과효천조인동혈청25-(OH)-VD 수평균저우정상대조조,차이유통계학의의(t=7.498,P <0.001)。VDR 기인서렬 rs7975232(ApaI)기인형 GG、GT,rs731236(TaqI)기인형 TT、TC,rs11574114기인형 GG、GC、CC,rs1544410(Bsm I)기인형 GG、GA, rs757343(Tru9I)기인형 GG、GA、AA,재량조간기인형적분포유명현적차이(P <0.01혹 P <0.05)。결론효천환인존재혈청25-(OH)-VD 불족혹결핍,VDR 기인다태위점 rs7975232、rs1544410、rs731236、rs11574114、rs757343기인형급등위기인분포재량조간존재차이,기다태성가능여효천발병상관。
Objective To explore the correlation of the 25-(OH)-VD and vitamin D receptor gene polymorphisms in children and asthma by detecting the level of 25-(OH)-VD and analyzing the gene polymorphisms of vitamin D receptor genes.Methods Forty children with asthma(clinical paracmasis)were enrolled as asthmatic group.Meanwhile,forty healthy children were selected as control group.Detecting the serum 25-(OH)-VD level in two groups children by the enzyme linked immunosorbent assay (ELISA ),meanwhile,examining the gene polymorphisms of vitamin D receptor gene rs2228570,rs7975232,rs1544410,rs731236,rs757343 by polymerase chain reaction (PCR),gene clone and DNA sequencing.Results The levels of 25-(OH)-VD in serum in asthma children were obviously lower than those in control children(t=7.498,P <0.001);the distributions of the genetypes GG and GT of vitamin D receptor gene rs7975232 (ApaI),the distributions of the genetypes TT and TC of rs731236 (TaqI),the distributions of the genetypes GG,GC and CC of rs11574114,the distributions of the genetypes GG and GA of rs1544410(Bsm I),the distributions of the genetypes GG,GA and AA of rs757343(Tru9I)in two groups children were all obviously different(P <0.01 or P <0.05).Conclusion There were deficiency or lack of 25-(OH)-VD in serum in asthma children,the distributions of vita-min D genetypes and allele of rs7975232,rs1544410,rs731236,rs11574114 and rs757343 were obviously different,the polymorphisms may be correlated to the pathogenesis of asthma.
