菊糖是一种聚果糖,平均分子量介于3000~5000d 之间。目前对菊糖的研究集中于其提取方法、性质和在食品行业的应用,对其进行疏水改性的研究较少。本实验以 N,N -二甲基甲酰胺( DMF)为溶剂,碳酸钾为碱性催化剂,通过转酯反应将月桂酸甲酯、肉豆蔻酸甲酯、棕榈酸甲酯以及油酸甲酯四种不同长度的脂肪酸链接枝到菊糖上,通过核磁共振氢谱验证产物结构,并对产物的理化性质及超声法制备的纳米粒的性质进行研究。结果表明,该合成方法能实现脂肪酸链的接枝以及菊糖的疏水改性;通过超声法可将产物制备成粒径100~300nm 的纳米粒,这些纳米粒可耐受胃液的酸性环境,常温下具有较好的放置稳定性,有望开发为疏水性纳米药物载体。

Inulin is a kind of polymeric fructose, whose average molecular weight ranges from 3000 to 5000d.Currently the research on inulin focus on its purification method , property and its application in the field of food science , while few attempt the hydrophobic modification of inulin .In this experiment, we succeed in grafting four kinds of fatty acids with different length , including methyl laurate, methyl myristate, methyl palmitate and Oil methyl ester, to the long chain of inulin through transesterification.In this case, Dimethylformamide ( DMF) serves as solvent and Potassium carbonate functions as catalyst.Then we demonstrate the structure of product with the approach of nuclear magnetic resonance and conduct reaserch on the property of the product as well as the nanoparticle produced by unltrasonic method .The result reveals that we can manage the grafting of fatty acid and the hydrophobic modification .And the nanoparticles whose diameter ranges from 100to 300 nanometer can tolerate the acidic environment in stomach and manifest quite good stability in room temperaure.Itˊs promised to be exploited as hydrophobic medical carrier .
