目的:通过双侧颈总动脉结扎法建立眼缺血综合征(OIS)大鼠实验动物模型。方法20只3月龄 BN 大鼠随机分为对照组(10只)与模型组(10只),模型组大鼠行双侧颈总动脉结扎建立 OIS 动物模型。观察大鼠造模术后1个月内的瞳孔对光反射情况;术后1个月行大鼠眼底彩色照相、眼底荧光血管造影(FFA)检查,然后处死大鼠获取视网膜,应用透射电镜检测视网膜结构变化。结果模型组中1只大鼠于术后第一天死亡,其余大鼠正常存活。模型组大鼠中3只双眼瞳孔对光反射消失,4只单眼瞳孔对光反射消失,2只双眼瞳孔对光反射均存在;与对照组相比,模型组大鼠大鼠出现眼底动脉硬化现象,FFA 的视网膜循环时间明显延长( P ﹤0.05);电镜结果显示,与正常对照组相比,模型组大鼠视网膜外核层疏松变薄、细胞超微结构紊乱呈凋亡表现。结论完全结扎大鼠双侧颈动脉可造成视网膜缺血以及形态学改变,可以尝试作为研究眼缺血综合征的动物模型。

목적:통과쌍측경총동맥결찰법건립안결혈종합정(OIS)대서실험동물모형。방법20지3월령 BN 대서수궤분위대조조(10지)여모형조(10지),모형조대서행쌍측경총동맥결찰건립 OIS 동물모형。관찰대서조모술후1개월내적동공대광반사정황;술후1개월행대서안저채색조상、안저형광혈관조영(FFA)검사,연후처사대서획취시망막,응용투사전경검측시망막결구변화。결과모형조중1지대서우술후제일천사망,기여대서정상존활。모형조대서중3지쌍안동공대광반사소실,4지단안동공대광반사소실,2지쌍안동공대광반사균존재;여대조조상비,모형조대서대서출현안저동맥경화현상,FFA 적시망막순배시간명현연장( P ﹤0.05);전경결과현시,여정상대조조상비,모형조대서시망막외핵층소송변박、세포초미결구문란정조망표현。결론완전결찰대서쌍측경동맥가조성시망막결혈이급형태학개변,가이상시작위연구안결혈종합정적동물모형。
Objective To establish rat model of ocular ischemic syndrome(OIS)by undergoing chronic bilateral common carotid artery occlusion(BCCAO). Methods We ligated the bilateral common carotid artery in the rat and established a model that is chronic bilateral common carotid artery occlusion(BCCAO). The study included 20 Brown Norway rats(BCCAO group:n = 10 rats;sham surgical group:n = 10). Both direct and consensual pupillary light reflexes were investigated before and after surgery,everyday for the first week and weekly for 30 days. At 1 months after baseline,rats underwent fundus color photography and fundus fluorescein angiography(FFA)examination. All rats were sacrificed and the structural changes of retina were observed by transmissiou electmn microscope. Results In the model group,1 rats died in the first day after surgery,and the other 9 rats survived. Three rats of the model group lost the direct pupillary reflex in both eyes,four rats lost this reflex in one eye,and two maintained it in both eyes. Compared to the sham group,the model group rats had fundus arteriosclerosis phenomenon,and the retinal circulation time of FFA was significantly longer in the model group( P ﹤ 0. 05). The results of electron microscopy showed that,compared with the normal control group,the retinal outer nuclear layer was loose and the cell ultrastructure was observed in the cell in the model group. Conclusion This study demonstrates that complete ligation of bilateral carotid artery can cause retinal ischemia and morphological changes,which might try as a research animal models of OIS.
