目的:检测糖尿病大鼠肝组织内脂素(visfatin)的表达,探讨内脂素在肝糖代谢的作用。方法雄性 SD 大鼠,分为5组:正常对照组(NC 组)、肥胖组(DIO 组)、糖尿病组(DM 组)、胰岛素治疗组(INS 组)、二甲双胍治疗组(MET 组)。检测大鼠空腹血糖(FPG)、总胆固醇(TC)、三酰甘油(TG)、游离脂肪酸(FFA )、空腹胰岛素(Fins)水平;RT‐PCR 测定大鼠肝组织 visfatin 、葡萄糖6磷酸酶(G‐6‐Pase)mRNA 的水平,Western blot 检测 visfatin 、磷酸化腺苷酸活化蛋白激酶‐α(p‐AMPKα)和腺苷酸活化蛋白激酶‐α(AMPKα)蛋白表达量。结果 DM 组 FBG 较 NC 组、DIO 组均显著升高(P<0.01);INS 组、MET 组 FBG 较 DM 组显著下降(P<0.01)。 DIO 组、DM 组 HOMA‐IR 均较 NC 组显著升高(P<0.01);DM 组 HOMA‐IR 较 DIO 组显著升高(P <0.01)。DIO 组、DM 组 ISI 均较 NC 显著降低(P<0.01),DM 组 ISI 较 DIO 组显著降低(P<0.01)。 DIO 组 TG 较 NC 组显著升高(P<0.01);INS 组、MET 组 TG 较 DM 组显著降低(P<0.05),DM 组、INS 组、MET 组 TG 、TC 均较 NC 组升高(P<0.05)。 DM 组、MET 组、INS 组 FFA 均较 NC 组明显升高(P<0.05);DM 组 FFA 较 DIO 组明显升高(P<0.05)。 DM 组 visfatin mRNA 表达较NC 组、DIO 组显著升高(P<0.05);INS 组、MET 组 visfatin mRNA 较 DM 组显著降低(P <0.01)。 DM 组、MET 、INS 组 G‐6‐Pase mRNA 较 DIO 组显著升高(P<0.05),MET 组 G‐6‐Pase mRNA 较 DM 组显著降低(P <0.05)。 DM 组、INS 组、MET 组visfatin 蛋白表达较 NC 组显著升高(P<0.05);DM 组、MET 组、INS 组 AMPKα蛋白表达较 NC 组显著降低(P<0.05),DM 组AMPKα蛋白表达较 DIO 组显著降低(P <0.05);DIO 组、DM 组、INS 组、MET 组 p‐AMPKα蛋白表达较 NC 组显著降低(P<0.01)。结论大鼠肝组织 visfatin 表达可能与肝糖脂代谢有关联,visfatin 可能未参与到二甲双胍激活 AMPK 降低血糖的机制中。
目的:檢測糖尿病大鼠肝組織內脂素(visfatin)的錶達,探討內脂素在肝糖代謝的作用。方法雄性 SD 大鼠,分為5組:正常對照組(NC 組)、肥胖組(DIO 組)、糖尿病組(DM 組)、胰島素治療組(INS 組)、二甲雙胍治療組(MET 組)。檢測大鼠空腹血糖(FPG)、總膽固醇(TC)、三酰甘油(TG)、遊離脂肪痠(FFA )、空腹胰島素(Fins)水平;RT‐PCR 測定大鼠肝組織 visfatin 、葡萄糖6燐痠酶(G‐6‐Pase)mRNA 的水平,Western blot 檢測 visfatin 、燐痠化腺苷痠活化蛋白激酶‐α(p‐AMPKα)和腺苷痠活化蛋白激酶‐α(AMPKα)蛋白錶達量。結果 DM 組 FBG 較 NC 組、DIO 組均顯著升高(P<0.01);INS 組、MET 組 FBG 較 DM 組顯著下降(P<0.01)。 DIO 組、DM 組 HOMA‐IR 均較 NC 組顯著升高(P<0.01);DM 組 HOMA‐IR 較 DIO 組顯著升高(P <0.01)。DIO 組、DM 組 ISI 均較 NC 顯著降低(P<0.01),DM 組 ISI 較 DIO 組顯著降低(P<0.01)。 DIO 組 TG 較 NC 組顯著升高(P<0.01);INS 組、MET 組 TG 較 DM 組顯著降低(P<0.05),DM 組、INS 組、MET 組 TG 、TC 均較 NC 組升高(P<0.05)。 DM 組、MET 組、INS 組 FFA 均較 NC 組明顯升高(P<0.05);DM 組 FFA 較 DIO 組明顯升高(P<0.05)。 DM 組 visfatin mRNA 錶達較NC 組、DIO 組顯著升高(P<0.05);INS 組、MET 組 visfatin mRNA 較 DM 組顯著降低(P <0.01)。 DM 組、MET 、INS 組 G‐6‐Pase mRNA 較 DIO 組顯著升高(P<0.05),MET 組 G‐6‐Pase mRNA 較 DM 組顯著降低(P <0.05)。 DM 組、INS 組、MET 組visfatin 蛋白錶達較 NC 組顯著升高(P<0.05);DM 組、MET 組、INS 組 AMPKα蛋白錶達較 NC 組顯著降低(P<0.05),DM 組AMPKα蛋白錶達較 DIO 組顯著降低(P <0.05);DIO 組、DM 組、INS 組、MET 組 p‐AMPKα蛋白錶達較 NC 組顯著降低(P<0.01)。結論大鼠肝組織 visfatin 錶達可能與肝糖脂代謝有關聯,visfatin 可能未參與到二甲雙胍激活 AMPK 降低血糖的機製中。
목적:검측당뇨병대서간조직내지소(visfatin)적표체,탐토내지소재간당대사적작용。방법웅성 SD 대서,분위5조:정상대조조(NC 조)、비반조(DIO 조)、당뇨병조(DM 조)、이도소치료조(INS 조)、이갑쌍고치료조(MET 조)。검측대서공복혈당(FPG)、총담고순(TC)、삼선감유(TG)、유리지방산(FFA )、공복이도소(Fins)수평;RT‐PCR 측정대서간조직 visfatin 、포도당6린산매(G‐6‐Pase)mRNA 적수평,Western blot 검측 visfatin 、린산화선감산활화단백격매‐α(p‐AMPKα)화선감산활화단백격매‐α(AMPKα)단백표체량。결과 DM 조 FBG 교 NC 조、DIO 조균현저승고(P<0.01);INS 조、MET 조 FBG 교 DM 조현저하강(P<0.01)。 DIO 조、DM 조 HOMA‐IR 균교 NC 조현저승고(P<0.01);DM 조 HOMA‐IR 교 DIO 조현저승고(P <0.01)。DIO 조、DM 조 ISI 균교 NC 현저강저(P<0.01),DM 조 ISI 교 DIO 조현저강저(P<0.01)。 DIO 조 TG 교 NC 조현저승고(P<0.01);INS 조、MET 조 TG 교 DM 조현저강저(P<0.05),DM 조、INS 조、MET 조 TG 、TC 균교 NC 조승고(P<0.05)。 DM 조、MET 조、INS 조 FFA 균교 NC 조명현승고(P<0.05);DM 조 FFA 교 DIO 조명현승고(P<0.05)。 DM 조 visfatin mRNA 표체교NC 조、DIO 조현저승고(P<0.05);INS 조、MET 조 visfatin mRNA 교 DM 조현저강저(P <0.01)。 DM 조、MET 、INS 조 G‐6‐Pase mRNA 교 DIO 조현저승고(P<0.05),MET 조 G‐6‐Pase mRNA 교 DM 조현저강저(P <0.05)。 DM 조、INS 조、MET 조visfatin 단백표체교 NC 조현저승고(P<0.05);DM 조、MET 조、INS 조 AMPKα단백표체교 NC 조현저강저(P<0.05),DM 조AMPKα단백표체교 DIO 조현저강저(P <0.05);DIO 조、DM 조、INS 조、MET 조 p‐AMPKα단백표체교 NC 조현저강저(P<0.01)。결론대서간조직 visfatin 표체가능여간당지대사유관련,visfatin 가능미삼여도이갑쌍고격활 AMPK 강저혈당적궤제중。
Objective To explore the effects of visfatin in glucose metabolism by testing the expression of visfatin in liver of rats in different glucose metabolic statuses .Methods SD rats were randomly divided into five groups :normal control group (NC group) ,diet induce obesity group (DIO group) ,diabetes mellitus group (DM group) ,diabetes controlled by insulin group (INS group)and diabetes controlled by metformin group (MET group) .Tested the data of blood glucose (FPG) ,triglyceride(TG) ,total cholesterol(TC) ,free fat acid(FFA) ,fasting insulin(Fins) .The liver of rats was used to test visfatin ,glucose‐6‐phosphatase(G‐6‐pase) mRNA by RT‐PCR and visfatin ,AMP‐activated protein kinase‐α (AMPKα) ,phosphor‐AMP‐activated protein kinase‐α (p‐AMPKα) protein by Western blot .Results FBG of group DM increased than group NC and DIO (P< 0 .01) ;FBG of group INS and MET decresed than group DM (P < 0 .01) ;HOMA‐IR of group DIO and DM increased than group NC (P < 0 .01) ;HOMA‐IR of group DM increased than group DIO (P< 0 .01) .ISI of group DIO and DM decresed than group NC (P< 0 .01) ;ISI of group DM de‐creased than group DIO(P< 0 .01) .TG of group DIO increased than group NC (P< 0 .01) .TG of group INS and MET decreased than group DM (P< 0 .05) .The level of TG and TC of group DM ,INS and MET increased than group NC (P< 0 .05) .The level of serum FFA of group DM ,INS and MET were significantly higher than group NC (P < 0 .05) ;FFA of group DM increased than group DIO(P< 0 .05) .The expression of visfatin mRNA of group DM increased than group NC and DIO (P< 0 .05) ;visfatin mRNA of group INS and MET decreased than group DM (P< 0 .01) .Group DM ,INS and MET had a significantly higher level of G‐6‐Pase mRNA of than group DIO( P < 0 .05) ;Group MET had a significantly lower level of G‐6‐Pase mRNA of than group DM (P <0 .05) .The expression of visfatin protein of group DM ,INS and MET increased than group NC ( P < 0 .05) .The expression of AMPKα protein of group DM ,INS and MET decresed than group NC(P< 0 .05) ;AMPKα of group DM decresed than group DIO (P<0 .05) .The expression of p‐AMPKα protein of group DIO ,DM ,INS and MET decresed than group NC (P< 0 .01) .Conclusion The ex‐pression of visfatin in liver of SD rats might have something to do with insulin resistance and diabetes .We could′t consider that visfatin can affect the pathway of metformin activated AMPK to decrease blood glucose .
