6 d龄紫花苜蓿(Mesicago sativa L.)幼苗在0、200 mmol/L NaCl处理9 d后,采用双向电泳分别分离了其根部蛋白组. 最终每块胶中有超过900个蛋白质点被分离,采用MALDI-TOF-TOF/MS质谱技术结合MASCOT软件在线检索,成功鉴定21个表达量发生1.5倍以上变化的蛋白质点,为19种不同的蛋白质. 生物信息学分析表明,这些差异表达蛋白质主要参与胁迫防御、碳水化合物和能量代谢、次生代谢、转录和翻译调控、信号传导和离子转运等调控途径.

6 day old alfalfa (Mesicago sativa L.) seedlings were exposed to 0(control),and 200 mmol/L NaCl for 9 days. Pro-teins extracted from the root of alfalfa seedlings were separated by two-dimensional gel electrophoresis(2-DE). More than 900 protein spots were detected on each gel,and 21 spots at least one point five-fold differences in abundance were identified by matrix-assisted laser desorption ionization time-of-flight/time-of-flight mass spectrometry (MALDI-TOF-TOF/MS) and MASCOT online. Bioinformatics analysis induced that these proteins were associated with a variety of functions,including stress defense, carbohydrate and energy metabolism,secondary metabolism,transcription and translation related,signaling and ion transport.
