为了满足种质资源保存、交换及杂交育种工作的需求,对小黑麦花粉活力及影响小黑麦花粉超低温保存效果的因素进行了研究.结果表明,采用TTC染色法测定小黑麦花粉的活力,结果相对准确,且其适合的浓度为0.1%;采用硅胶干燥处理3 h后,小黑麦花药相对含水量为34.5%,比较适合超低温保存,且保存前后花粉活力保持稳定,其活力相对保持率为92.3%;超低温冷冻保存5 d后,采用不同的化冻方式进行化冻,结果发现自来水冲洗化冻后小黑麦花粉活力相对较高,说明自来水冲洗化冻是简便适合的化冻方式.

In order to meet the demands of germplasm resources preservation, exchange and cross breeding, triticale pollen vitality and the cryopreservation-influencing factors were studied in this research. The results showed that, TTC staining method was relatively accurate for determining the triticale pollen vitality, and the suitable concentration was 0.1%; more-over, it was more suitable for cryopreservation when relative water content of anther reached to 34.5% after dried three hours by silica gel, which made pollen vitality stable and its vitality relative retention rate was 92.3%.In addition, different ways of defrosting were studied after five days of cryopreservation, and the results showed that the pollen vitality was relatively high when adopting method of tap water washing, indicating that tap water defrosting was more simple and applicable.
