目的:利用细胞原代及传代培养技术将大鼠骨髓基质干细胞诱导分化为成骨细胞和脂肪细胞,为其进一步应用奠定基础。方法全骨髓法分离大鼠骨髓基质干细胞,传代后分别在成骨、成脂诱导条件下继续培养,采用碱性磷酸酶染色、茜素红染色及油红“O”染色观察其成骨及成脂分化结果。结果第2代大鼠骨髓基质干细胞成骨诱导9 d后碱性磷酸酶染色呈阳性细胞,连续诱导14 d后可见矿化结节形成,成脂诱导14 d后可见脂肪细胞形成。结论随着诱导条件的不同,大鼠骨髓基质干细胞在体外可定向分化为成骨细胞和脂肪细胞。
目的:利用細胞原代及傳代培養技術將大鼠骨髓基質榦細胞誘導分化為成骨細胞和脂肪細胞,為其進一步應用奠定基礎。方法全骨髓法分離大鼠骨髓基質榦細胞,傳代後分彆在成骨、成脂誘導條件下繼續培養,採用堿性燐痠酶染色、茜素紅染色及油紅“O”染色觀察其成骨及成脂分化結果。結果第2代大鼠骨髓基質榦細胞成骨誘導9 d後堿性燐痠酶染色呈暘性細胞,連續誘導14 d後可見礦化結節形成,成脂誘導14 d後可見脂肪細胞形成。結論隨著誘導條件的不同,大鼠骨髓基質榦細胞在體外可定嚮分化為成骨細胞和脂肪細胞。
목적:이용세포원대급전대배양기술장대서골수기질간세포유도분화위성골세포화지방세포,위기진일보응용전정기출。방법전골수법분리대서골수기질간세포,전대후분별재성골、성지유도조건하계속배양,채용감성린산매염색、천소홍염색급유홍“O”염색관찰기성골급성지분화결과。결과제2대대서골수기질간세포성골유도9 d후감성린산매염색정양성세포,련속유도14 d후가견광화결절형성,성지유도14 d후가견지방세포형성。결론수착유도조건적불동,대서골수기질간세포재체외가정향분화위성골세포화지방세포。
Objective To classify rat bone marrow stromal cells into lipoblast and osteoblasts in vitro by using corned cells and subculture, and to provide a foundation for its further application. Method The rat bone marrow stromal cells were separated by whole bone marrow adherent screening method. The cells were cultured in lipoblast and osteoblasts induced conditions after propagated generations. The results of differentiation was observed by alkaline phosphatase staining,alizarin red staining and oil red “O” staining. Results After 9 days of continuous induction,positive cells stained by alkaline phosphatase could be observed. After 14 days,calcified nodules and adipocytes formed in different situations, respectively. Conclusion The rat bone marrow stromal cells can differentiate into osteoblasts and adipocytes under different induction conditions.
