目的:探讨单核苷酸多态性位点rs6551328多态性与黑龙江地区汉族女性乳腺癌发病相关性,及其基因型分布与乳腺癌风险的关系。方法采用病例-对照研究策略,采用PCR-Sequencing研究方法对150例黑龙江地区汉族女性乳腺癌及300例年龄性别相匹配健康对照进行分析,分析rs6551328等位基因在病例-对照中的分布差异,并比较基因型分布与乳腺癌发病风险及临床病理特征的关系。结果 rs6551328 C/T两个等位基因分布在两组中未检测到差异,而CC,CT,TT基因型分布差异有统计学意义(P<0.05),CT基因型相对于CC基因型,风险度OR=1.776,95%CI为1.155~2.728;CT杂合基因型相对于纯合基因型(CC+TT),风险度OR=1.883,95%CI为1.195~2.281。根据临床资料进一步分析,携带CT基因型乳腺癌转移风险增加。结论 rs6551328多态性位点与黑龙江地区汉族女性乳腺癌发病相关,其中基因型CT是乳腺癌发病易感基因型,且该基因型与乳腺癌转移相关。

Objective To explore the correlation of Loci rs 6551328 polymorphism and the attack of Breast Cancer in fe-male of Han group in Heilongjiang province. Methods 150 breast cancer patients and 300 cases of age matched healthy controls without breast cancer family history were recruited from clinic. General clinical data and 5ml peripheral venous blood from each cases were collected. Genomic DNA was extracted by kit. PCR/sequencing was used for genotyping. Allele frequency and genotype frequency was analyzed for comparison in both groups. The relationship of rs6551328 allele geno-type distribution with risk of Breast cancer and clinical pathological features was studied by statistical analysis. Results There was no significant difference between two groups on Allele gene C/T frequency distribution, while CC,CT,TT genotype frequency distribution showed statistical difference(P<0.05). Compared with genotype CC, OR of CT was 1.776, with 95%CI from 1.155~2.278. Compared with homozygous genotype (CC+TT), OR of heterozygous genotype CT was 1.883,with 95%CI from 1.195 to 2.281. Combined with clinic information, the risk of tumor metastasis was detected increased in patients with CT genotype. Conclusion Loci rs 6551328 polymorphism is correlated with the attack of Breast Cancer in female of Han group in Heilongjiang province. The CT genotype is the susceptible genotype, which related to tumor metastasis.
