应用差分脉冲伏安法、循环伏安法、控制电位电解法等电化学方法和紫外光谱法,研究了中药提取物鹿蹄草素在玻碳电极上的电化学行为及其与DNA的相互作用,并考察其相关电化学动力学参数。结果表明,鹿蹄草素在玻璃电极上发生了两个电子转移的电极反应,并且主要是受吸附控制的表面过程,其在玻碳电极上的电子转移数为2;随着DNA的加入,鹿蹄草素的峰电流降低、峰电位发生正移,说明鹿蹄草素与DNA通过嵌插方式相互作用,生成复合物,根据加入DNA后使鹿蹄草素的峰电流变化动力学参数,也可定量分析检测DNA的浓度。

The electrochemical behaviors of Pyrolin as well as its interaction with DNA were studied using cyclic voltammetry, differential pulse voltammetry, controlled potential electrolysis and ultraviolet-visible spectroscopy. The results showed that the electrode process of Pyrolin on glassy carbon electrode was two electrons and two hydrions transfered,the surface is mainly controlled by absorption process,and the elec-tron transfer coefficient was 2. In the presence of DNA,the oxidation current of Pyrolin was decreased con-siderably,and the peak potential was shifted to the positive direction,which suggested that the binding of Pyrolin to DNA was via intercalation. According to the concentration of the reduce peak current after the addition of DNA,determined the quantitative detection of the concentration of DNA,respectively.
