目的:运用脱氢表雄酮(DHEA)诱导 SD 大鼠多囊卵巢综合征模型,研究模型大鼠的生殖表型和代谢表型。方法选取23日龄雌性 SD 大鼠,模型组每日颈背部皮下注射 DHEA 6 mg/100 g 体重,对照组每日颈背部皮下注射0.2 ml 注射油剂。注射20 d 后观察两组大鼠动情周期、卵巢形态学、性激素、体重、脂肪湿重、卵巢体积、OGTT 血糖、空腹胰岛素、胰岛素抵抗指数(HOMA - IR)及血脂等生殖及代谢相关指标的变化。结果模型组大鼠失去规律的动情周期;卵巢体积增大,呈多囊样变;血清促卵泡生成素(FSH)、促黄体生成素(LH)、睾酮(T)水平明显高于对照组;糖耐量异常,空腹胰岛素及 HOMA 指数明显高于对照组;血清胆固醇、脂肪湿重、脂体比明显高于对照组。结论 DHEA 可诱导 SD 大鼠出现与 PCOS 患者相似的生殖和代谢异常的改变,是研究多囊卵巢综合征生殖及代谢异常的较理想的动物模型。

목적:운용탈경표웅동(DHEA)유도 SD 대서다낭란소종합정모형,연구모형대서적생식표형화대사표형。방법선취23일령자성 SD 대서,모형조매일경배부피하주사 DHEA 6 mg/100 g 체중,대조조매일경배부피하주사0.2 ml 주사유제。주사20 d 후관찰량조대서동정주기、란소형태학、성격소、체중、지방습중、란소체적、OGTT 혈당、공복이도소、이도소저항지수(HOMA - IR)급혈지등생식급대사상관지표적변화。결과모형조대서실거규률적동정주기;란소체적증대,정다낭양변;혈청촉란포생성소(FSH)、촉황체생성소(LH)、고동(T)수평명현고우대조조;당내량이상,공복이도소급 HOMA 지수명현고우대조조;혈청담고순、지방습중、지체비명현고우대조조。결론 DHEA 가유도 SD 대서출현여 PCOS 환자상사적생식화대사이상적개변,시연구다낭란소종합정생식급대사이상적교이상적동물모형。
Objective To induce SD rat model of polycystic ovarian syndrome( PCOS)with de-hydroephiandrosterone(DHEA)and study the reproductive and metabolic phenotypes in model rats. Methods The female SD rats,aged 23 days were selected. In the model group,the subcutaneous injection of DHEA was done at the dorsal neck region,6mg/ 100g body mass. In the control group,the subcutaneous injection of oil was done at the dorsal neck region. The relevant reproductive and metabolic indexes were observed in 20 days of injection in the two groups,such as estrous cycle,ovarian morphology,sexual hormone,body weight,wet weight of fat,ovarian volume,OGTT,fasting blood insulin,HOMA - IR and blood lipid. Results In the mod-el group,the regular estrous cycle was lost,ovarian volume was increased and polycystic changes occurred. The serum FSH,LH and T levels were apparently higher than those in the control group. The glucose toler-ance abnormality and the levels of fasting insulin and HOMA index were apparently higher than those in the control group. The serum cholesterol level,fat wet weight and lipid proportion were apparently higher than those in the control group. Conclusion DHEA can induce SD rat to have the similar reproductive and meta-bolic abnormality as PCOS patients. It is the satisfactory approach to the preparation of animal model for the study of reproductive and metabolic abnormality of PCOS.
