目的：探讨益气活血药(生脉注射液联合血塞通注射液)对心肌梗死大鼠心室缝隙连接蛋白43(Cx43)表达及室颤发生的影响。方法采用结扎心脏左冠状动脉前降支的方法建立心肌梗死大鼠模型，另设假手术(只穿线，不结扎)作对照。术后随机分为模型组、假手术组、卡托普利组和益气活血组。卡托普利组给予卡托普利片4.39 mg/(kg· d)溶解灌胃；益气活血组给予生脉注射液3.51 mL/(kg· d)，血塞通注射液17.54 mg/(kg· d)腹腔注射；模型组和假手术组给予生理盐水，疗程4周。治疗结束后取材，采用 real time PCR法检测左心室梗死边缘区 Cx43 mRNA的表达；Western blot法检测 Cx43蛋白表达；在体电刺激记录室颤诱发率。结果与假手术组比较，模型组 Cx43的 mRNA和蛋白表达均显著降低(P<0.05或P<0.01)；与模型组比较，卡托普利组和益气活血组 Cx43的 mRNA和蛋白表达均显著升高(P<0.05或P<0.01)。在体电刺激室颤诱发率模型组最高，卡托普利组和益气活血组的室颤诱发率降低，差异有统计学意义(P<0.05)。结论生脉注射液联合血塞通注射液可以减少心肌梗死后室颤的发生，其机制与改善心室 Cx43表达有关。
목적：탐토익기활혈약(생맥주사액연합혈새통주사액)대심기경사대서심실봉극련접단백43(Cx43)표체급실전발생적영향。방법채용결찰심장좌관상동맥전강지적방법건립심기경사대서모형，령설가수술(지천선，불결찰)작대조。술후수궤분위모형조、가수술조、잡탁보리조화익기활혈조。잡탁보리조급여잡탁보리편4.39 mg/(kg· d)용해관위；익기활혈조급여생맥주사액3.51 mL/(kg· d)，혈새통주사액17.54 mg/(kg· d)복강주사；모형조화가수술조급여생리염수，료정4주。치료결속후취재，채용 real time PCR법검측좌심실경사변연구 Cx43 mRNA적표체；Western blot법검측 Cx43단백표체；재체전자격기록실전유발솔。결과여가수술조비교，모형조 Cx43적 mRNA화단백표체균현저강저(P<0.05혹P<0.01)；여모형조비교，잡탁보리조화익기활혈조 Cx43적 mRNA화단백표체균현저승고(P<0.05혹P<0.01)。재체전자격실전유발솔모형조최고，잡탁보리조화익기활혈조적실전유발솔강저，차이유통계학의의(P<0.05)。결론생맥주사액연합혈새통주사액가이감소심기경사후실전적발생，기궤제여개선심실 Cx43표체유관。
Objective To study the effects of Yiqi Huoxue drugs(YQHX,combine Shengmai injection with Xuesaitong injection)on the expression of connexin 43(Cx43)and ventricular fibril ation in rats with myocardial infarction.Methods Model rats of myocardial in-farction were established by occlusion of the left anterior descending coronary artery,the sham operation group was without occlu-sion.After operation rats were randomly divided into sham operation group,model group,YQHX group and captopril group. YQHX [Shengmai injection 3.51 mL/(kg· d)and Xuesaitong injection 17.54 mg/(kg· d)]were administered intraperitoneal y.Captopril[4.39 mg/(kg· d)]and distil ed water (for the sham operation and model groups)were administered oral y.The course of treatment was 4 weeks.The expressions of Cx43 mRNA of myocardium in left ventricular infarct border zone were detected by real time fluorescent quantitative polymerase chain reaction after treatment.The expression of Cx43 protein was observed by western blot.The incidence of ventricular fibril ation was recorded by electric stimulation in vivo.Results Compared with sham operation group,the expressions of Cx43 mRNA and protein decreased remarkably in the model group (P<0.05 or P<0.01).Compared with model group,the expres-sions of Cx43 mRNA and protein increased significantly in YQHX group and captopril group (P<0.05 or P<0.01).The incidence of ventricular fibril ation was the highest in model group.The incidence of ventricular fibril ation reduced significantly in captopril group and YQHX group(P<0.05).Conclusion Shengmai injection and Xuesaitong injection can reduce the occurrence of ventricular fibril-lation after myocardial infarction.It is related to improving the expressions of Cx43.
