目的:探讨环磷酰胺(CTX )诱导膀胱肿瘤的机制。方法 SD大鼠12只,随机分为两组,处理组和对照组各6只。处理组大鼠给与环磷酰胺100 mg/kg腹腔注射,对照组给与同等剂量的生理盐水腹腔注射;24 h后取膀胱,采用HE染色观察形态结构的变化,采用免疫组织化学方法检测KI67、P53、P21的表达定位,采用Western blot的方法检测KI67、P53、P21的蛋白表达量,并进行数据统计。结果 CTX处理组膀胱出现明显的水肿、出血、粘连;HE染色可见,膀胱黏膜弯曲减少、肿胀,细胞排列疏松,膀胱黏膜呈现毛刺样改变;免疫组织化学染色可见KI67/P53/P21在膀胱黏膜上皮细胞表达;Western blot检测及数据统计可知,KI67蛋白在CTX组表达高于对照组(P<0.05);P53/P21蛋白表达在CTX组低于对照组(P<0.05)。结论环磷酰胺可降低膀胱黏膜P53/P21蛋白的表达,诱导膀胱肿瘤的发生。

목적:탐토배린선알(CTX )유도방광종류적궤제。방법 SD대서12지,수궤분위량조,처리조화대조조각6지。처리조대서급여배린선알100 mg/kg복강주사,대조조급여동등제량적생리염수복강주사;24 h후취방광,채용HE염색관찰형태결구적변화,채용면역조직화학방법검측KI67、P53、P21적표체정위,채용Western blot적방법검측KI67、P53、P21적단백표체량,병진행수거통계。결과 CTX처리조방광출현명현적수종、출혈、점련;HE염색가견,방광점막만곡감소、종창,세포배렬소송,방광점막정현모자양개변;면역조직화학염색가견KI67/P53/P21재방광점막상피세포표체;Western blot검측급수거통계가지,KI67단백재CTX조표체고우대조조(P<0.05);P53/P21단백표체재CTX조저우대조조(P<0.05)。결론배린선알가강저방광점막P53/P21단백적표체,유도방광종류적발생。
ABSTRACT:Objective To explore the mechanism of cyclophosphamide induced bladder tumor .Methods A total of 12 SD rats were randomly divided into two groups:treatment group (n= 6) and control group (n= 6) .Rats in the treatment group received intraperitoneal injection of cyclophosphamide 100 mg/kg ,and rats in the control group received the same dose of intraperitoneal injection of saline .After 24 hours ,the bladders were harvested to observe the morphological changes .HE staining was used to observe the change of morphological structure ,immunohistochemical method was adopted to detect the ex‐pression of KI67 ,P53 and P21 ,and Western blot was used to detect the protein expression of KI67 ,P53 and P21 .Results The treatment group showed visible edema ,hemorrhage and adhesion .HE staining showed less bladder mucosa bending ,swell‐ing ,loosely arranged cells ,and burr change .Immunohistochemical staining detected KI67/P53 ,P21 expression in bladder mu‐cosa .Western showed the treamnet group had higher KI67 protein expression (P<0 .05);while the control group had higher P53 /P21 protein expression (P<0 .05) .Conclusion Cyclophosphamide can induce bladder tumor by reducing the expres‐sion of P53 /P21 protein .
