目的:观察不同剂量右美托咪定预处理对大鼠全脑缺血再灌注损伤炎性反应的影响。方法全脑缺血前2 h,假手术组(S 组),对照组(C 组)静注生理盐水2 ml/ h,低剂量右美托咪定预处理组(D1组)、高剂量右美托咪定预处理组(D2组)分别静注右美托咪定每分钟0.05μg/ kg和每分钟0.5μg/ kg。全脑缺血再灌后30分钟检测血清肿瘤坏死因子(TNF)-α和白细胞介素(IL)-1β变化;测定脑组织含水量;观察海马 CA1区神经病理变化。结果与 C 组比较,D1组、D2组TNF-α、IL-1β水平显著减少( P ﹤0.05),D1组、D2组之间比较差异无统计学意义(P ﹥0.05)。D1组、D2组较 C 组固缩神经元数量及神经元缺失显著减少。结论全脑缺血再灌注损伤大鼠,低、高剂量右美托咪定预处理均可产生一定的脑保护作用。

목적:관찰불동제량우미탁미정예처리대대서전뇌결혈재관주손상염성반응적영향。방법전뇌결혈전2 h,가수술조(S 조),대조조(C 조)정주생리염수2 ml/ h,저제량우미탁미정예처리조(D1조)、고제량우미탁미정예처리조(D2조)분별정주우미탁미정매분종0.05μg/ kg화매분종0.5μg/ kg。전뇌결혈재관후30분종검측혈청종류배사인자(TNF)-α화백세포개소(IL)-1β변화;측정뇌조직함수량;관찰해마 CA1구신경병리변화。결과여 C 조비교,D1조、D2조TNF-α、IL-1β수평현저감소( P ﹤0.05),D1조、D2조지간비교차이무통계학의의(P ﹥0.05)。D1조、D2조교 C 조고축신경원수량급신경원결실현저감소。결론전뇌결혈재관주손상대서,저、고제량우미탁미정예처리균가산생일정적뇌보호작용。
Objective To observe the influence of different doses of dexmedetomidine pretreat-ment on inflammatory reaction after ischemic-reperfusion injury in rats. Methods Two hours before the ischemia,dexmedetomidine was given in low dosage group[group D1,0. 05μg/( kg·min)]and high dosage[group D2,0. 05μg/(kg·min)],while normal saline(2 ml/ h)was given instead of dexmedetomi-dine in the sham operation group( group S)and controls( group C). Blood samples were taken for the measurement of TNF-αand IL-1β at the 30th minute after reperfusion. Water content of the brain tissues and pathological changes in the hippocampal CA1 area were also observed. Results Compared with group C,the levels of TNF-α and IL-1β were significantly decreased in group D1 and D2(P ﹥ 0. 05),but no sig-nificant difference was observed between group D1 and group D2. Compared with group C,the number of shrinkage neuronal cell and neuron loss in group D1 and D2 was decreased. Conclusion Dexmedetomi-dine pretreatment can produce certain brain protective effects in rats with global cerebral ischemia-reperfu-sion injury.
