目的:研究星形胶质细胞上调基因‐1(AEG‐1)、β‐连环蛋白(β‐catenin)和C‐myc在原发性肝癌组织中的表达情况,以探讨AEG‐1、β‐catenin、C‐myc蛋白在肝癌发生、发展中的可能作用。方法以40例原发性肝癌患者术后肝癌组织及癌旁组织作为研究组,8例正常肝组织标本作为对照组,采用免疫组织化学染色及western blot方法测定各组研究对象中AEG‐1、β‐catenin及C‐myc蛋白的表达水平。结果在肝癌组织中AEG‐1、β‐catenin和C‐myc蛋白的表达水平显著高于癌旁组织及正常肝组织,其组间差异有统计学意义(P<0.05);在肝癌组织中AEG‐1蛋白和β‐catenin蛋白的表达呈正相关(r=0.420),β‐catenin蛋白和C‐myc蛋白的表达呈正相关(r=0.388),AEG‐1蛋白和C‐myc蛋白的表达呈正相关(r=0.332),P<0.05。结论 AEG‐1、β‐catenin和C‐myc蛋白在肝癌组织中的表达水平明显升高,提示AEG‐1可能通过Wnt信号传导途径调控下游的β‐catenin和C‐myc蛋白,促进肿瘤细胞增殖从而参与肝细胞肝癌的发生发展。

목적:연구성형효질세포상조기인‐1(AEG‐1)、β‐련배단백(β‐catenin)화C‐myc재원발성간암조직중적표체정황,이탐토AEG‐1、β‐catenin、C‐myc단백재간암발생、발전중적가능작용。방법이40례원발성간암환자술후간암조직급암방조직작위연구조,8례정상간조직표본작위대조조,채용면역조직화학염색급western blot방법측정각조연구대상중AEG‐1、β‐catenin급C‐myc단백적표체수평。결과재간암조직중AEG‐1、β‐catenin화C‐myc단백적표체수평현저고우암방조직급정상간조직,기조간차이유통계학의의(P<0.05);재간암조직중AEG‐1단백화β‐catenin단백적표체정정상관(r=0.420),β‐catenin단백화C‐myc단백적표체정정상관(r=0.388),AEG‐1단백화C‐myc단백적표체정정상관(r=0.332),P<0.05。결론 AEG‐1、β‐catenin화C‐myc단백재간암조직중적표체수평명현승고,제시AEG‐1가능통과Wnt신호전도도경조공하유적β‐catenin화C‐myc단백,촉진종류세포증식종이삼여간세포간암적발생발전。
Objective To determine the expression of the astrocyte‐elevated gene‐1 (AEG‐1) ,β‐catenin and C‐myc in hepatocellular carcinoma (HCC) .It will provide better evidence to investigate their significance in the onco‐genesis and progression .Method Hepatocarcinomous tissues and adjacent tissues were collected from forty postoper‐ative HCC patients ,and then collected eight normal liver tissue (NT) samples .The expressions level of AEG‐1 ,β‐catenin and C‐myc in hepatocarcinomous tissues (HCTs) and adjacent tissues (ATs) were tested via Immunohisto‐chemical (IHC) assay ,as well as the Western blotting (WB) assay in each sample .Result The expressions level of AEG‐1 ,β‐catenin and C‐myc in HCTs were significantly higher in ATs and NTs .Comparisons between groups all showed statistical significance (P< 0 .05) .Relative expression quantities in HCTs between AEG‐1 and β‐catenin demonstrated positive correlation (r=0 .420) ,relative expression quantities in HCTs between β‐catenin and C‐myc demonstrated positive correlation (r= 0 .388) ,relative expression quantities in HCTs between AEG‐1 and C‐myc demonstrated positive correlation (r=0 .332) ,and were statistically significant (P<0 .05) .Conclusion The expres‐sion levels of AEG‐1 ,β‐catenin and C‐myc in human hepatocellular carcinoma tissues elevated significantly ,which indicates that AEG‐1 may involve in downstream proteins of β‐catenin and C‐myc in Wnt signal pathway ,to facilitate the development of HCC by promoting proliferation of cancer cells .
