目的:采用中药提取浓缩机组从知母中提取总皂苷,并对其含量进行测定。方法取知母饮片适量,加8倍量75%乙醇,回流提取2次,每次1.5h,收集浓缩滤液。经AB-8或D-101大孔吸附树脂分离、纯化,冷冻干燥后制成知母总皂苷提取物粉末。提取物经乙醇和盐酸混合物水解成萨尔萨皂苷元,用氯仿萃取;萨尔萨皂苷元经浓硫酸氧化、显色后,紫外分光光度法法测定光密度值,计算总皂苷的纯度。结果得到3批知母总皂苷提取物粉末,得药率依次为2.6%、6.2%和6.8%,纯度分别为65.3%、59.1%和61.4%。结论中药提取浓缩机组提取知母总皂苷容量大且得率高,2种大孔吸附树脂分离知母总皂苷效果相似。

목적:채용중약제취농축궤조종지모중제취총조감,병대기함량진행측정。방법취지모음편괄량,가8배량75%을순,회류제취2차,매차1.5h,수집농축려액。경AB-8혹D-101대공흡부수지분리、순화,냉동간조후제성지모총조감제취물분말。제취물경을순화염산혼합물수해성살이살조감원,용록방췌취;살이살조감원경농류산양화、현색후,자외분광광도법법측정광밀도치,계산총조감적순도。결과득도3비지모총조감제취물분말,득약솔의차위2.6%、6.2%화6.8%,순도분별위65.3%、59.1%화61.4%。결론중약제취농축궤조제취지모총조감용량대차득솔고,2충대공흡부수지분리지모총조감효과상사。
Objective To extract total saponin from a Chinese herb Rhizoma Anemarrhenae, and determine its con-tent.Methods A small amount of decoction pieces of Rhizoma Anemarrhenae was placed in its eight times of volumes of 75%ethanol and then subject to reflux extraction for twice, 1.5 hours for each time.The concentrated filtrate was collect-ed and separated through AB-8 or D-101 macroporous resin.The resultant product was purified before lyophilization into total saponin powders.The obtained extract was hydrolyzed by the mixture of ethanol and hydrochloric acid into sar-sasapogenin, which was then extracted with chloroform.The absorption of sarsasapogenin was determined by ultraviolet spectrophotometry after sulfuric acidoxidation and development.Then purity of total saponin was calculated.Results Three batches of timosaponin powders were obtained.The yield was 2.6%, 6.2%, and 6.8%, respectively.The purity was 65.3%, 59.1%, and 61.4%, respectively.Conclusion Timosaponins can be extracted from Rhizoma Anemarrhe-nae, with a high yield, which can be separated by the two types of macroporous resin used in a similar manner.
