目的:探讨高良姜素对口腔癌CAL27细胞的生长和转移的影响。方法采用酶抑制动力学方法观察高良姜素对基质金属蛋白酶9(MMP-9)的抑制作用,分析不同浓度高良姜素对口腔癌CAL27细胞的毒理性、转移的抑制作用和CAL27细胞中 TIMP-1、MMP-9、转化生长因子( TGF-β1)表达水平的影响。结果高良姜素能够抑制MMP-9的活性,诱导CAL27细胞的凋亡,但对正常细胞的毒性较低。随着高良姜素浓度的增加,口腔癌CAL27细胞中MMP-9和TGF-β1表达均呈降低趋势,TIMP-1表达呈升高趋势,且口腔癌CAL27细胞穿过滤膜的能力下降。结论高良姜素可通过下调MMP-9和TGF-β1的水平、增强TIMP-1表达,抑制口腔癌CAL27细胞的生长和转移。

목적:탐토고량강소대구강암CAL27세포적생장화전이적영향。방법채용매억제동역학방법관찰고량강소대기질금속단백매9(MMP-9)적억제작용,분석불동농도고량강소대구강암CAL27세포적독이성、전이적억제작용화CAL27세포중 TIMP-1、MMP-9、전화생장인자( TGF-β1)표체수평적영향。결과고량강소능구억제MMP-9적활성,유도CAL27세포적조망,단대정상세포적독성교저。수착고량강소농도적증가,구강암CAL27세포중MMP-9화TGF-β1표체균정강저추세,TIMP-1표체정승고추세,차구강암CAL27세포천과려막적능력하강。결론고량강소가통과하조MMP-9화TGF-β1적수평、증강TIMP-1표체,억제구강암CAL27세포적생장화전이。
Objective To investigate the effect of Galangin on CAL27 cell growth and metastasis of patients with oral carcinoma. Methods The inhibitory effect of Galangin on matrix metalloproteinase-9 (MMP-9) was studied using enzyme inhibition kinetics method, and the effects of Galangin with different concentrations on the inhibition of CAL27 cell growth and metastasis and on expressions of tissue inhibitor of metalloproteinase (TIMP-1), MMP-9 and transforming growth factor-β1 (TGF-β1) in CAL27 cells were also analyzed. Results Galangin could inhibit MMP-9 activity, induce CAL27 apoptosis with little toxicity for the normal cells. With the increasing concentration of Galangin, MMP-9 and TGF-β1 expression levels in CAL27 oral carcinoma cells were decreasing, and TIMP-1 expression level was increasing, and the ability of CAL27 oral carcinoma cells through artificial basement membrane was degraded. Conclusion Galangine may enhance TIMP-1 expression by inhibiting the MMP-9 and TGF-β1 levels, eventually playing an inhibitory role on CAL27 cell growth and metastasis.
