目的:探讨荧光原位杂交( FISH)检测乳腺癌表皮生长因子受体2( HER2)基因扩增在淮海经济区的推广和应用。方法通过组织芯片技术,运用免疫组织化学( IHC)法和FISH分别检测124例浸润性乳腺癌组织中HER2蛋白表达和HER2基因扩增状况,并进行对比分析。结果124例乳腺癌中HER2蛋白表达为(眓眓眓)39例,(眓眓)70例,(0,+)15例,其FISH阳性率分别为87.18%、24.29%、20.00%,FISH法与IHC法检测结果总体符合率为85.19%。结论在淮海经济区,采用IHC检测Her2蛋白表达为(眓眓)时与Her2基因扩增状态存在较大差异,IHC呈阳性患者有必要选择性地进行FISH检测,有助于指导临床治疗及判断患者预后。

Objective To discuss the promotion and application of fluorescence in situ hybridization ( FISH) to de-tect the amplification of human epithelial growth factor receptor 2 (HER2) gene in breast cancer patients of Huaihai eco-nomic zone.Methods A total of 124 human breast cancer samples were prepared by tissue microarray ( TMA) .Their expression of HER2 at protein and mRNA levels was determined by immunohistochemistry ( IHC) and FISH.Then, both methods were compared for their consistency.Results Among the samples were 39 HER2 strongly positive samples (艹+), 70 positive ones (艹), and 15 weak positive or negative ones (0/+), according to IHC staining.Using FISH, the corresponding positive rates of HER2 gene were 87.18%, 24.29%, and 20.00%respectively, which showed 85.19%of consistence with IHC results.Conclusion There is a big difference between the expression of HER2 protein (艹) de-tected by IHC and gene amplification in the Huaihai economic zone.HER2 positive carriers are necessary to undergo FISH in order to improve clinical treatment and prognosis.
