目的:建立一种双黄连粉针剂中黄芩苷的含量测定方法,用于控制双黄连粉针剂的质量.方法:采用岛津LC-20A高效液相色谱仪和Shim-Pack C18色谱柱(150mm ×4.6mm I.D.,5μm);流动相为乙腈-0.5%冰醋酸水溶液,梯度洗脱;流速1.0 mL/min;二极管阵列检测,检测波长为280 nm;柱温:30℃.结果:方法的专属性良好,黄芩苷在浓度 4μg/mL ~20μg/mL 范围内,线性关系良好(r =0.9998),日内精密度RSD=0.24%,n=5,日间精密度RSD=0.43%,n=3,黄芩苷的定量限为2μg/mL,检测限为0.67μg/mL,平均回收率100.23%,回收率RSD=1.95%,n=9.结论:建立了一种专属性强,精密度高,回收率良好的用于双黄亮粉针剂中黄芩苷含量测定的高效液相色谱分析方法.

Objective:To develop an quantitative HPLC method for the determination of baicalin in Shuanghuan-glian powder for injection.Methods: The HPLC analysis was carried out on LC-20A HPLC and a Shim-Pack C18 column(150 mm ×4.6mm I.D.,5μm) was used, with a gradient elution of acetonitrile-0.5%gla-cial acetic acid solution at the flow rate of 1.0mL/min.DAD was detected and the detective wave length was set at 280nm, and the column temperature was at 30℃.Results:The specificity of this method was good,and the calibration curve was linear within the range of 4~20μg/mL and for baicalin ( r=0.9998) .The intra-day precision was RSD 0.24%, n=5.The inter-day precision was RSD 0.43%, n=3.The Quantitative limit of baicalin was 2μg/mL,and the detection limit of baicalin was 0.67μg/mL.The average recovery for the baicalin was 100.23%(RSD=1.95%).Conclusion:With a good specificity, high precision and well re-covery the HPLC method was developed to determine the baicalin in Shuanghuanglian powder for injection.
