目的 研究四君子汤复方多糖对荷瘤小鼠免疫功能的影响.方法 建立小鼠S180实体瘤模型,分别给予低、中、高剂量四君子复方多糖,测定各组小鼠肿瘤生长抑制率、脾脏指数和胸腺指数;采用MTT法测定荷瘤小鼠脾淋巴细胞增殖作用;流式细胞技术(FACS)检测脾细胞表面抗原CD4、CD8的表达;酶联免疫吸附实验法(ELISE)测定小鼠血清白细胞介素-2(IL-2)γ-干扰素(IFN-γ)含量.结果 中、高剂量的四君子汤复方多糖对S180肉瘤的抑制率分别为20.26%和23.28%,与模型组比较有显著性差异(P<0.05);中、高剂量的四君子汤复方多糖可以显著提高荷瘤小鼠的脾脏指数和胸腺指数;模型对照组小鼠CD4/CD8比值较正常小鼠明显下降,而四君子汤复方多糖可以显著提高荷瘤小鼠CD4/CD8比值;四君子汤复方多糖实验组小鼠血清IL-2和IFN-γ的含量都显著高于模型组小鼠(P<0.05).结论 四君子汤复方多糖具有显著的免疫增强活性,可以改善荷瘤小鼠的免疫损伤状况,发挥一定的抑瘤作用.

목적 연구사군자탕복방다당대하류소서면역공능적영향.방법 건립소서S180실체류모형,분별급여저、중、고제량사군자복방다당,측정각조소서종류생장억제솔、비장지수화흉선지수;채용MTT법측정하류소서비림파세포증식작용;류식세포기술(FACS)검측비세포표면항원CD4、CD8적표체;매련면역흡부실험법(ELISE)측정소서혈청백세포개소-2(IL-2)γ-간우소(IFN-γ)함량.결과 중、고제량적사군자탕복방다당대S180육류적억제솔분별위20.26%화23.28%,여모형조비교유현저성차이(P<0.05);중、고제량적사군자탕복방다당가이현저제고하류소서적비장지수화흉선지수;모형대조조소서CD4/CD8비치교정상소서명현하강,이사군자탕복방다당가이현저제고하류소서CD4/CD8비치;사군자탕복방다당실험조소서혈청IL-2화IFN-γ적함량도현저고우모형조소서(P<0.05).결론 사군자탕복방다당구유현저적면역증강활성,가이개선하류소서적면역손상상황,발휘일정적억류작용.
Obective To study immunological Effect of total polysaccharides from Sijunzi decoction (SJP) in tumor-bearing mice. Methods S180 tumor-bearing mice models were established and were given three different doses of SJP. The tumor growth inhibitory rate, spleen index and thymus index were calculated. MTT assay was used to observe the effect of SJP on spleen lymphocyes in vitro separated from tumor-bearing mice. The expression of surface antigens CD4 and CD8 in the spleen were detected by flow cytometry technique. The contents of IL-2 and IFN-γ were determined by means of ELISA.Results The tumor inhibition rates of medium and high-dose SJP were 20.26% and 23.28%, significantly compared with the model groups. Spleen index and thymus index were significantly increased in medium and high-dose SJP groups.The ratio of CD4/CD8 in model group was lower than in normal group, SJP could up-regulated markedly CD4/CD8 value in tumor-bearing mice. The levels of IL-2 and IFN-γ in serum from SJP-treated mice, increased significantly compared to model control group(P< 0.05).Conclusion SJP had strong immunostimulating activity and could improve immunological damage in tumor-bearing mice.
