目的:探讨Prohibitn2(PHB2)在正常和损伤脊髓组织中的表达变化.方法:将56只SD大鼠随机分为6个实验组和1个对照组,实验组按Allen法制作急性脊髓损伤模型,并分别按伤后6h、1d、3d、5d、7d和14d取材.用Western blot和免疫组织化学法观察PHB2在正常和损伤大鼠脊髓组织中的表达情况.结果:PHB2在大鼠正常脊髓灰质和白质中均有表达,且分布较均匀,PHB2蛋白表达从脊髓损伤后6h开始迅速下降,持续至3d,之后逐渐上升,于2w后接近正常水平.结论:PHB2在脊髓灰质和白质中均有表达,并在脊髓损伤后表达发生变化,提示PHB2在脊髓损伤中过程中起到一定作用.

Objective:To investigate expression of Prohibitn2 (PHB2) in normal and injured spinal cords. Methods:56 SD rats were randomly divided into six experimental groups and the control group. In the experimental groups, the spinal cord injury (SCI) model was made according to the Allen's method, and the animals were killed at 6h,1d, 3d, 5d, 7d and 14d after injury. The expression of PHB2 in normal and injured spinal cord tissue was observed by Western blot and immunohistochemical methods. Results: PHB2 was localized in both gray matter and white matter in normal rat spinal cords. The expression of PHB2 decreased at 6th h after spinal cord injury, continued to 3th d, then increased gradually, and become close to the normal level at 14th d. Conclusion: PHB2 was localized in both gray and white matter of spinal cords, and changed in expression after spinal cord injury, suggesting that PHB2 might be involved in the regeneration or degeneration of the injured spinal cord.
