目的 观察洛铂与放疗联合治疗对SGC-7901胃癌细胞凋亡的影响.方法 收集功能正常的SGC-7901胃癌细胞与正常的胃壁细胞做为研究对象.使用统计学随机分组的原则,将SGC-7901胃癌细胞划分为单纯化疗组、单纯放疗组以及放疗化疗联合组,将正常的胃壁细胞作为对照组,随后试验用的SGC-7901胃癌细胞进行功能的检查,随后将化疗组的SGC 7901胃癌细胞进行化疗治疗,对单纯放疗组的胃癌细胞使用放疗治疗,对放疗化疗联合组进行洛铂与放疗联合治疗,然后通过使用显微镜观察各种细胞生长状态与生长速度,然后使用流式细胞仪测量正常细胞与SGC-7901胃癌细胞的凋亡率,最后测量p53 mRNA表达程度.结果 通过显微镜对细胞组织的形态进行观察,普通的细胞组织体现为多角形状,细胞体积基本一致.联合应用洛铂和照射之后,观察到细胞增长显著缓慢,细胞形态缩小,发生明显的细胞增长抑制.通过检查细胞的死亡率发现单纯化疗组和单纯放疗组的细胞死亡率都明显多于正常对照组细胞.同时联合组胃癌细胞的死亡率明显多于单纯化疗组和单纯放疗组,同时洛铂与放疗联合治疗组的p53 mRNA表达程度显著多于对照组.结论 通过洛铂与放疗联合使用对SGC-7901胃癌细胞可以明显的提高细胞的死亡率.

목적 관찰락박여방료연합치료대SGC-7901위암세포조망적영향.방법 수집공능정상적SGC-7901위암세포여정상적위벽세포주위연구대상.사용통계학수궤분조적원칙,장SGC-7901위암세포화분위단순화료조、단순방료조이급방료화료연합조,장정상적위벽세포작위대조조,수후시험용적SGC-7901위암세포진행공능적검사,수후장화료조적SGC 7901위암세포진행화료치료,대단순방료조적위암세포사용방료치료,대방료화료연합조진행락박여방료연합치료,연후통과사용현미경관찰각충세포생장상태여생장속도,연후사용류식세포의측량정상세포여SGC-7901위암세포적조망솔,최후측량p53 mRNA표체정도.결과 통과현미경대세포조직적형태진행관찰,보통적세포조직체현위다각형상,세포체적기본일치.연합응용락박화조사지후,관찰도세포증장현저완만,세포형태축소,발생명현적세포증장억제.통과검사세포적사망솔발현단순화료조화단순방료조적세포사망솔도명현다우정상대조조세포.동시연합조위암세포적사망솔명현다우단순화료조화단순방료조,동시락박여방료연합치료조적p53 mRNA표체정도현저다우대조조.결론 통과락박여방료연합사용대SGC-7901위암세포가이명현적제고세포적사망솔.
Objective To observe the effect of lobaplatin combined with radiotherapy on apoptosis of SGC-7901 gastric cancer cell.Methods The SGC 7901 gastric cancer cells with normal function and normal cells were collected as the objects of study.SGC-7901 gastric cancer cells were divided into gastric cancer chemotherapy group, radiotherapy group and radiotherapy combined with chemotherapy group.The normal gastric parietal cells were as the control group, then tests for SGC-7901 gastric cancer cell function examination, then the SGC 7901 gastric cancer cells in chemotherapy group were given chemotherapy treatment, the radiotherapy treatment were given for gastric cancer cells in radiotherapy group, Lobaplatin combined with radiotherapy were given for the treatment of gastric cancer cells in chemotherapy combined with radiotherapy group, and then the cell growth states and growth rates were observed by microscope, and then use flow cytometry to measure the apoptosis of normal cells and SGC-7901 gastric cancer cell rate, and the degree of expression of p53 mRNA was measured.Results Through the observation of morphology of tissue cells of ordinary microscope, reflects the multi angle shape, the cell volume was basically the same.After the combination of lobaplatin and radiation, cell growth was slow, the cell morphology was significantly reduced, cell growth was inhibited.The cell death of chemotherapy group and radiotherapy group were significantly higher than those of normal controls.At the same time, the mortality rate of gastric cancer cells in combination group was significantly more than that of the chemotherapy group and the radiotherapy group, and the P53 mRNA expression level in the lobaplatin combined with radiotherapy treatment group was significantly higher than that in the normal control group.Conclusions Lobaplatin combined with radiotherapy can significantly improve the cell death rate of gastric cancer cell line SGC-7901.
