目的 优选金蝉花中麦角甾醇的最佳提取工艺. 方法 选择乙醇浓度、提取温度和提取时间为自变量,麦角甾醇提取量为因变量,在单因素试验基础上,采用星点设计-效应面法优选提取工艺,通过Design-Expert 8. 0. 6统计分析软件对试验数据进行多元线性模型和二次多项式模型拟合,并绘制效应面图和等高线图.结果 金蝉花中麦角甾醇的最佳提取工艺为以88%乙醇为提取溶剂,在74 ℃下提取38 min;麦角甾醇检测浓度在9. 28~148. 50 μg/mL范围内与其峰面积呈良好的线性关系,平均加样回收率为97. 99%. 结论 优化的条件简单、快速,可为金蝉花的进一步开发利用和有效性研究提供理论依据.

Objective To optimize the extraction process of ergosterol in cordyceps cicadae. Methods Based on single factor tests,central composite design-response surface methodology was applied to optimize the process with ethanol concentration,extraction temperature and extraction time as independent variables,and the extraction amounts of ergosterol as dependent variable. Design-Expert 8. 0. 6 statistic analysis software was used to fit multivariate linear equa-tion and second-order polynomial equation for experimental data, and delineate response surface and contour plot. Results The best technical condition was as follows:the extraction lasted for 38 min under 74 ℃ with 88% alco-hol as the extration solvent. The linear range of ergosterol was 9. 28~148. 50 μg/mL and the average recovery rate was 97. 99%. Conclusion The method is simple and rapid,and it provides scientific basis for the safety and effectiveness of cordyceps cicadae in clinical use.
