目的 观察雷公藤甲素对膝部骨关节炎滑膜c-Jun、MMP-9及血清PGE2、IL-8水平的影响,探讨其在膝部骨关节炎中的药理学价值. 方法 45只SD大鼠随机分为正常对照组、模型对照组及雷公藤甲素组,建立碘乙酸钠(Monosodium iodoacetate,MIA)膝关节炎模型,并给予雷公藤甲素干预,以 Western blot 及(qRT)-PCR法检测c-Jun及MMP-9在膝关节滑膜中蛋白及mRNA水平表达,以酶联免疫法检测外周血中PGE2 及IL-8水平. 结果 Western blot及( qRT)-PCR显示,c-Jun及MMP-9在膝关节滑膜组织中蛋白及mRNA水平上均有表达,雷公藤甲素组及正常对照组明显低于模型对照组(P<0. 01),雷公藤甲素组与正常对照组比较差异无统计学意义(P>0. 05);在血清PGE2 及IL-8表达上,雷公藤甲素组及正常对照组明显低于模型对照组(P<0. 05).结论 雷公藤甲素可抑制骨关节炎滑膜中c-Jun及MMP-9表达,降低外周血PGE2 及IL-8水平,抑制炎症反应,对骨关节炎有治疗作用.

목적 관찰뢰공등갑소대슬부골관절염활막c-Jun、MMP-9급혈청PGE2、IL-8수평적영향,탐토기재슬부골관절염중적약이학개치. 방법 45지SD대서수궤분위정상대조조、모형대조조급뢰공등갑소조,건립전을산납(Monosodium iodoacetate,MIA)슬관절염모형,병급여뢰공등갑소간예,이 Western blot 급(qRT)-PCR법검측c-Jun급MMP-9재슬관절활막중단백급mRNA수평표체,이매련면역법검측외주혈중PGE2 급IL-8수평. 결과 Western blot급( qRT)-PCR현시,c-Jun급MMP-9재슬관절활막조직중단백급mRNA수평상균유표체,뢰공등갑소조급정상대조조명현저우모형대조조(P<0. 01),뢰공등갑소조여정상대조조비교차이무통계학의의(P>0. 05);재혈청PGE2 급IL-8표체상,뢰공등갑소조급정상대조조명현저우모형대조조(P<0. 05).결론 뢰공등갑소가억제골관절염활막중c-Jun급MMP-9표체,강저외주혈PGE2 급IL-8수평,억제염증반응,대골관절염유치료작용.
Objective To detect the effect of triptolide on c-Jun and MMP-9 in knee osteoarthritis synovium and serum PGE2 and IL-8,discuss the pharmacological value of triptolide. Methods Forty-five SD rats were divided into normal control group,model control group and triptolide group randomly,MIA osteoarthritis models were construc-ted,and triptolide was given. The c-Jun and MMP-9 expression at protein and mRNA level in knee synovium were de-tected by Western blot and ( qRT)-PCR. Results Western blot and ( qRT)-PCR showed that c-Jun and MMP-9 were expressed at protein and mRNA levels in synovium tissues,the expression in triptolide group and normal control group was higher than those of model control group (P<0. 01),there was no significant difference between triptolide group and normal control group (P>0. 05). Serum PGE2 and IL-8 in triptolide group and normal control group were lower than those of model control group (P<0. 01),there was no significant difference between triptolide group and normal control group (P>0. 05). Conclusion Triptolide can inhibit c-Jun and MMP-9 expression in osteoarthritis synovium, decrease the PGE2 and IL-8 levels in peripheral blood,inhibit the inflammatory reaction,it has therapeutic effect on os-teoarthritis.
