目的 建立大鼠心肌缺血/再灌注损伤模型,观察红景天苷对急性心肌缺血/再灌注损伤大鼠心功能的影响. 方法 将10只未造模的正常大鼠作为对照组,其余30只大鼠随机分为缺血/再灌注模型组、红景天苷组、红景天苷+普萘洛尔组并造模. 于造模成功24 h后迅速断头取心,采用Langendorff离体心脏灌流方法,分别灌流红景天苷、红景天苷+普萘洛尔,测定给药前及给药后1、4h急性心肌缺血/再灌注损伤离体心脏的心率、心肌收缩张力、左室最大上升/下降速率( ± LVdp/dtmax )、主动脉流量( AF)、冠脉流量( CF). 结果 给药后,红景天苷组的心肌收缩张力、± LVdp/dtmax、AF及CF明显升高,与对照组、缺血/再灌注模型组比较,差异有统计学意义(P<0. 05);红景天苷+普萘洛尔组心肌收缩张力、± LVdp/dtmax、AF及CF先上升后下降,与对照组、缺血/再灌注模型组比较,差异有统计学意义( P<0. 05 ). 结论 红景天苷对离体急性心肌缺血/再灌注损伤大鼠离体心脏具有正性肌力作用,β受体阻断剂普萘洛尔可以阻断这种强心效应,提示其对β受体有激动作用,可增强心肌收缩力,提高心输出量,增加冠脉流量,进而改善急性缺血心脏心功能.

목적 건립대서심기결혈/재관주손상모형,관찰홍경천감대급성심기결혈/재관주손상대서심공능적영향. 방법 장10지미조모적정상대서작위대조조,기여30지대서수궤분위결혈/재관주모형조、홍경천감조、홍경천감+보내락이조병조모. 우조모성공24 h후신속단두취심,채용Langendorff리체심장관류방법,분별관류홍경천감、홍경천감+보내락이,측정급약전급급약후1、4h급성심기결혈/재관주손상리체심장적심솔、심기수축장력、좌실최대상승/하강속솔( ± LVdp/dtmax )、주동맥류량( AF)、관맥류량( CF). 결과 급약후,홍경천감조적심기수축장력、± LVdp/dtmax、AF급CF명현승고,여대조조、결혈/재관주모형조비교,차이유통계학의의(P<0. 05);홍경천감+보내락이조심기수축장력、± LVdp/dtmax、AF급CF선상승후하강,여대조조、결혈/재관주모형조비교,차이유통계학의의( P<0. 05 ). 결론 홍경천감대리체급성심기결혈/재관주손상대서리체심장구유정성기력작용,β수체조단제보내락이가이조단저충강심효응,제시기대β수체유격동작용,가증강심기수축력,제고심수출량,증가관맥류량,진이개선급성결혈심장심공능.
Objective To establish the rat model of myocardial ischemia/reperfusion injury,and observe the effects of salidroside on cardiac function of rats with acute myocardial ischemia/reperfusion injury. Methods Forty rats were divided into 4 groups:control group (n=10),ischemia/reperfusion model group (model group),salidroside group,salidroside+ propranolol group. The rats were sacrificed at 24 h after successful modeling,and the hearts were quickly isolated from the rats. Rats in salidroside group,salidroside+propranolol group were perfused with salidroside and salidroside+propranolol. The heart rate,myocardial contraction tension,left ventricular maximum increase/decrease rate ( LVdp/dtmax ) ,aortic blood flow ( AF) ,coronary flow ( CF) of isolated hearts in rats with acute myocardial ische-mia/reperfusion injury were determined. Results After medication,the myocardial contractile tension, ± LVdp/dtmax , AF and CF in salidroside group increased significantly,and the indexes in salidroside+propranolol group increased at first and then decreased, there were significant differences ( salidroside group vs. control group, salidroside group vs. model group,salidroside+propranolol group vs. control group,salidroside+propranolol group vs. model group,P<0. 05). Conclusion Salidroside has positive inotropic effect on heart of rat with acute myocardial ischemia/reperfusion injury in vitro,which could be blocked by propranolol. It is indicated that salidroside could enhance myocardial contractili-ty,improve the cardiac output and heart function in acute myocardial ischemia.
