Lilium brownii var .viridulum%Scales%Differentiation
以平陆食用百合脱毒试管苗的鳞片为外植体 ,研究了不同接种部位和方式 ,不同植物激素种类和浓度对鳞片分化的影响.结果表明 :平陆百合鳞片的分化能力为外层>中层>内层 ,同一鳞片的不同部位分化能力为下段>中段>上段.接种方式以鳞片竖直插入培养基中为好 ,鳞片分化快 ,分化率高.试验中设置的培养基均可诱导鳞片分化 ,但附加不同种类和浓度的激素对鳞片分化出的小鳞茎的数量和生长状况影响不同 ,差异很大.其中以培养基MS+6-BA 0.8 mg·L -1 +NAA 0.1 mg·L -1上诱导出的小鳞茎数量多 ,生长壮 ,适宜作为快繁时获取大量鳞茎的培养基 ;在培养基MS+ZT 1.0 mg·L -1 +NAA 0.2 mg·L -1和MS+ZT 1.5 mg·L -1 + IBA 0.1 mg·L -1上诱导出的小鳞茎数量不多 ,但生长健壮 ,可直接成苗 ,适宜作为诱导鳞片分化较大鳞茎的培养基 ;诱导鳞片分化愈伤组织的最适宜培养基为MS+2 ,4-D 1.0 mg·L -1 +6-BA 0.5 mg·L -1 ,愈伤组织不经转换培养基可直接分化鳞茎.

Using the test tube flake seedings of Lilium brownii var .viridulum as explants ,we studied different inocula-tion sites and methods ,the effect of different plant hormones species and concentrations on the differentiation of scales . The results showed that :The differentiation ability of Scale in Lilium brownii var .viridulum was :Outer > middle> inner layer ,the ability to different parts of the same scale was :Upper> middle> section and the vertical scale in-serts the culture medium ,the differentiation and differentiation rats were the best .The Experiment seted all of the me-dium that could induce the differentiation of scales .The number and growth status of the small bulbs which were dif-ferentiated from the scales were differed greatly ,because of the different kinds and concentrations of hormones .MS+6-BA 0.8 mg·L -1 +NAA 0.1 mg·L -1 was induced the large number of small bulbs ,growth and strong ,the medium was suitable for the development of a large number of bulbs in the rapid propagation .The number of small bulbs in-duced by MS+ZT 1.0 mg·L -1 +NAA 0.2 mg·L -1 and MS+ZT 1.5 mg·L -1 +IBA 0.1 mg·L -1 was not much ,but strong greatly and directly seeding ,suitable as a medium for inducing the differentiation of the larger bulbs .The most suitable medium was MS+2 ,4-D1.0 mg·L -1 +6-BA 0.5 mg·L -1 which induced scale differentiation of callus ,the cal-lus without conversion medium direct differentiation of the bulb .
