为改善骨胶原的营养和生理功能 ,优化羊骨粉的碱性蛋白酶水解工艺 ,制备高水解度的胶原多肽 ,以邻苯二甲醛法测定水解度 ,在单因素试验基础上 ,采用响应面分析法 ,建立了水解度与料水比(X1 )、温度(X2 )、时间(X3 )、加酶量(X4 )的二次多项回归数学模型 ,方差分析结果表明此模型拟合度高.对模型最优解适当调整 ,X1、X2、X3、X4分别为9% (W/V)、45 ℃ 、325 min和4.25% (E/S) ,pH9条件下酶解 ,测得水解度为28.12% ,接近预测值28.39% ,进一步说明所建模型可靠 ,可对不同条件下的水解度进行预测 ,为羊骨的高值化、安全化利用提供理论依据.

To improve the nutritional and physiological function of bone collagen ,parameters were optimized to prepare ossein peptides with high degree of hydrolysis (DH) by hydrolyzing sheep bone powder with the protease of alcalase . DH was determined by o-phthalaldehyde(OPA) methodology .In light of the results of single-factor experiments ,re-sponse surface analysis was carried out and a mathematical model was established to describe the DH in response to the levels of bone powder concentration (X1 ) ,temperature(X2 ) ,time(X3 ) ,and the amount of added alcalase .Variance a-nalysis suggested that the model showed high fitting degree .Sheep bone powder was hydrolyzed at pH 9 and under the adjusted optimized parameters of the model ,X1 、X2 、X3 、X4 was 9% (W/V)、45 ℃ 、325 min and 4.25% (E/S)respec-tively ,the determined DH was 28.12% ,which was close to the predicted 28.39% .This further confirmed the reliabili-ty of the established model which can be used to predict the DH when sheep bone powder was hydrolyzed by alcalase . The results of this study will also provide a theoretical guidance for the high-value and safe utilization of sheep bone .
