目的:用异硫氰酸荧光素(FITC)对人血清白蛋白(HSA)进行荧光标记。方法:采用直接标记法,并考查不同投料比时的荧光素分子与蛋白分子的结合情况、荧光标记物(FITC-HSA)的荧光光谱及荧光寿命情况。结果:FITC-HSA的激发波长为500 nm,荧光波长为530 nm;其固态和液态低温保存时荧光寿命分别为一年和37天。
目的:用異硫氰痠熒光素(FITC)對人血清白蛋白(HSA)進行熒光標記。方法:採用直接標記法,併攷查不同投料比時的熒光素分子與蛋白分子的結閤情況、熒光標記物(FITC-HSA)的熒光光譜及熒光壽命情況。結果:FITC-HSA的激髮波長為500 nm,熒光波長為530 nm;其固態和液態低溫保存時熒光壽命分彆為一年和37天。
목적:용이류청산형광소(FITC)대인혈청백단백(HSA)진행형광표기。방법:채용직접표기법,병고사불동투료비시적형광소분자여단백분자적결합정황、형광표기물(FITC-HSA)적형광광보급형광수명정황。결과:FITC-HSA적격발파장위500 nm,형광파장위530 nm;기고태화액태저온보존시형광수명분별위일년화37천。
OBJECTIVE:To label human serum albumin(HSA).METHODS:In this study,we labeled human serum albumin with fluorescein isothiocyanate(FITC),and then the ratio of FITC to HSA for labeling reaction,the varances of spectra after being labeled and the life of the labeled HSA were optimized.RESULTS:The result showed that the spectra were λex=500 nm,λem=530 nm;the life of the labeled HSA were 37 day and 1 year in liquid and solid,respectively.CONCLUSION:To label HSA with FTIC was successful,and it was applicable to Fluoroimmunoassay.
