目的检测系统性红斑狼疮(SLE)患者血浆白细胞介素(IL)-35水平,探讨其在SLE患者血浆中的变化及与疾病活动性的关系。方法采用酶联免疫吸附试验(ELISA)检测30例SLE患者及15名健康体检者血浆IL-35水平,比较其在各组中的差异及其与SLE患者部分临床指标的相关性,两组间比较用独立样本t检验,三组间比较采用方差分析,与临床资料的相关分析采用非参数的spearmen的相关分析。结果活动期SLE患者血浆IL-35水平(10.7±3.9)pg/ml低于健康对照组(55.2±15.6)pg/ml (P<0.01)及SLE稳定组(25.3±8.1)pg/ml (P<0.05),稳定组亦低于健康对照组(P<0.05)。SLE患者血浆IL-35水平与抗核抗体(ANA)、抗dsDNA、红细胞沉降率(ESR)、24h尿蛋白定量、SLE活动指数(SLEDAI)呈负相关,与补体(C)3呈正相关。结论 SLE患者血浆IL-35水平降低,与疾病活动性相关,提示IL-35在SLE的进展中可能发挥重要作用,并可作为反映SLE活动性的指标。

Objective To investigate the serum IL-35 levels in patients with systemic lupus erytbematosus (SLE), explore the role of IL-35 in the pathogenesis of SLE and their relationship with disease activity. Methods 30 hospitalized SLE patients were recruited and 15 healthy volunteers were enrolled as normal controls, serum IL-35 levels in the peripheral blood of SLE patients and healthy controls were measured by enzyme-linked immunosorbent assay (ELISA). Independent sample t test was used for statistical analysis between two groups, analysis of variance (ANOVA) was applied for data analysis between three groups,non-parameterical Spearman’s analysis was used for correlation analysis. Results The serum IL-35 levels was decreased in active SLE patients [(10.7±3.9) pg/ml)] compared with inactive SLE patients [(25.3±8.1) pg/ml (P<0.05)] and healthy controls [(55.2±15.6) pg/ml) (P<0.01)], inactive SLE patients is lower than controls (P<0.05) , the serum IL-35 levels were negatively correlated with the titer of anti-dsDNA and ANA、SLEDAI、levels of 24-hour urinary protein and ESR, positively correlated with the levels of C3. Conclusion The serum IL-35 levels in SLE patients were elevated significantly and correlated with disease activity, these results demonstrate IL-35 may play an important role in the pathogenesis of SLE, the serum IL-35 levels are parameters for evaluation of SLE activity and severity.
