目的建立以反相高效液相色谱法测定人血浆中血药浓度地西泮的方法。方法采用Diamonsil C18(250nm×4.6nm，5μm)色谱柱，流动相为甲醇：水(65：35)，流速为1.0ml/min，检测波长为254nm，柱温为25℃，采用蛋白沉淀法处理血浆样品。结果地西泮血浆浓度在5.15～25.75μg(r =0.9991)范围内线形关系良好，低、中、高三种浓度的平均相对回收率分别为103.01％、101.87％，99.09％，RSD均小于4％。结论本方法简便、快速、准确、灵敏度高，可用于地西泮血药浓度测定和人体药动学研究。

Objective:To establish a Rp-HPLC method for the determination of diazepam in human plasma.Method:The HPLc separation was achieved on a Diamonsil C18 column(250mm×4.6 mm,5μm),and the mobile phase consisted of MeOH-H20(65:35).The detection wavelength wan 254 nm.flow rate was 1.0ml/L.and column temperature was25℃. The protein precipitation method was used to treat the plasma samples.Result:The linear range of diazepam was 5.15~25.75μg (r=0.9995),the average relative recovery of low,middle and high concentration was103.01%,101.87%and 99.09%,respectively,and the RSDs were less than 4%.Conclusion:The method iS simple,rapid,accurate and sensitive for the plasma concentration monitoring and clinical pharmacokinetic studies of diazepam.
