目的观察类胰蛋白酶对肝星状细胞(HSC)Ⅰ型胶原和蛋白水解酶激活受体-2(PAR-2)表达的影响,探讨类胰蛋白酶在肝纤维化中的作用.方法用Percoll密度梯度离心法分离、培养大鼠HSC,经系列浓度类胰蛋白酶作用后,应用RT-PCR法检测Ⅰ型胶原和PAR-2mRNA 的表达.结果类胰蛋白酶(1~100ng/ ml)能够促进HSCⅠ型胶原及PAR-2mRNA的表达,此作用呈剂量依赖性(各实验组与对照组比较,P <0.05).结论类胰蛋白酶可能通过PAR-2作用于HSC,促进HSC表达Ⅰ型胶原,参与肝纤维化的发生发展过程.

Objective To observe the effects of tryptase on the expression of collagen I and PAR-2 in hepatic stellate cell (HSC)and to study the role played by tryptase during development of liver fibrosis. Methods HSC were isolated from rat liver with percoll and treated with an increasing amount of tryptase. The expression of collagen I and protease-activated receptor-2(PAR-2) mRNAs in vitro cultured HSC was assessed by means of semi-quantitive reverse transcriptase polymerase chain reaction (RT- PCR). Results Tryptase (1~100ng/ml) promoted expression of collagen I mRNA and PAR-2 mRNA of HSC in a dose-dependent manner (P<0.05). Conclusion Tryptase may promote the collagen I expression of HSC by PAR-2 imposed on HSC which might be involved in pathogenesis and development of liver fibrosis.
