GABAAR%GABAARα1mRNA%Cerebral ischemia
目的观察大鼠大脑皮质γ-氨基丁酸A受体(GABAAR)及受体基因(GABAARα1mRNA)在脑缺血不同时段的表达及其动态变化的规律,探讨其在急性脑缺血中变化的意义.方法55只雄性Wistar大鼠随机分为对照组,脑缺血6 h组、12 h组、24 h组、3 d组、7 d组(均为手术组),假手术组;采用大鼠大脑中动脉栓塞(MCAO)模型,至规定时间点取大脑组织,用光学显微镜观察神经元损伤程度,用形态分析系统检测各组G A B A A R表达的阳性细胞数.用地高辛标记寡聚核苷酸探针原位杂交技术与计算机分析系统在光学显微镜下观察GABAARα1mRNA在大脑皮质阳性神经元的表达情况.结果各手术组GABAAR及GABAARα1mRNA表达均低于假手术组及对照组,与对照组相比, GABAAR在缺血6 h其表达既有降低,至12 h降至最低.GABAARα1mRNA表达在缺血24h开始下降,3d下降显著,7d恢复正常,而假手术组与对照组相比无统计学意义.结论 GABAAR及 GABAARα1mRNA在急性脑缺血时含量降低可能为脑缺血时脑损伤的重要原因.

Objecive To observe the expression of GABAAR and GABAARα1mRNA and the dynamic regulation in the cortex of rats with focal cerebral ischemia in different phases and to investigate its significance in acute ischemia. Methods: Fifty-five male Wistar rats were randomly divided into 11 groups: The normal control, sham operation and ischemic operation group(subdivided into 6,12,24 hour and 3,7 days) by middle cerebral artery occlusion model. Brain specimens of the rats from each group were obtained to observe the damage of neurons with light microscope and to judge the positive expression of neurons in immunohistochemistry technique, and then to determine the positive cell number ex-pressed by GABAAR in each group with morphologic analysis system, and the changes of GABAARα1mRNA expression were observed under light microscope with digoxin labeled oligonucleotide probe in situhybridization method and computer aided image analysis. Results: Compared with the normal control group and the sham operation group, the expression of GABAAR and GABAARα1mRNA in each ischemia operation group was decreased. the expression of GABAAR was decreased after 6h and reached the bottom within 12 hours after ischemia. the expression of GABAARα1mRNA was decreased from 24h and was the lowest within 3d and recovered to normal in 7d after ischemia. However, there was no significant difference between the normal control group and sham operation group. Conclusion:It is suggested that the decreasing of content of GABAAR and GABAARα1mRNA after ischemia might be related with ischemia- injury.
