目的通过检测胃癌患者血清和胃癌组织中8-OHdG的水平,探究8-羟基脱氧鸟苷在胃癌的发生和发展中的作用。方法 选取2010年7月至2011年1月泰安市中心医院住院均经病理确诊的胃癌患者30例,慢性胃炎患者15例。正常对照20例,来自健康人群志愿者。用酶联免疫吸附法(ELISA)测定研究对象血清中的8-OHdG的水平,同时采用免疫组化SP法检测研究对象胃组织中8-OHdG水平,并分别对胃癌患者与正常对照组血清8-OHdG水平以及胃癌患者与慢性胃炎及正常对照组胃组织8-OHdG表达情况进行比较。结果 胃癌组织中8-OHdG的表达状况与胃癌组织的组织分化程度密切相关,在高中分化腺癌中,8-OHdG的表达情况明显低于低分化的腺癌(p<0.05)。患者胃癌组织中8-OHdG的表达与胃癌患者的性别、年龄无明显相关(p>0.05)。结论 8-OHdG的水平变化可能与胃癌的发生发展有关,提示血清8-OHdG可能为辅助胃癌诊断的重要血清学标志物。8-OHdG与胃癌组织的分化程度、临床分期、浸润胃壁的深度以及是否伴有淋巴结转移等状况密切有关,可作为病理活检中辅助标志物判断胃癌病变。

목적통과검측위암환자혈청화위암조직중8-OHdG적수평,탐구8-간기탈양조감재위암적발생화발전중적작용。방법 선취2010년7월지2011년1월태안시중심의원주원균경병리학진적위암환자30례,만성위염환자15례。정상대조20례,래자건강인군지원자。용매련면역흡부법(ELISA)측정연구대상혈청중적8-OHdG적수평,동시채용면역조화SP법검측연구대상위조직중8-OHdG수평,병분별대위암환자여정상대조조혈청8-OHdG수평이급위암환자여만성위염급정상대조조위조직8-OHdG표체정황진행비교。결과 위암조직중8-OHdG적표체상황여위암조직적조직분화정도밀절상관,재고중분화선암중,8-OHdG적표체정황명현저우저분화적선암(p<0.05)。환자위암조직중8-OHdG적표체여위암환자적성별、년령무명현상관(p>0.05)。결론 8-OHdG적수평변화가능여위암적발생발전유관,제시혈청8-OHdG가능위보조위암진단적중요혈청학표지물。8-OHdG여위암조직적분화정도、림상분기、침윤위벽적심도이급시부반유림파결전이등상황밀절유관,가작위병리활검중보조표지물판단위암병변。
Aims: we detected the serum and tissue expression of 8-OHdG in patients with gastric carcinoma. The aim of our study was to reveal the role that 8-oxo-7, 8-dihydro-2'-deoxyguanosine (8-OHdG) played in the development of gastric carcinoma.Methods: 30 patients with gastric carcinoma, 15 with chronic gastritis and 20 healthy control subjects were included from July 2010 to January 2011 in Tai An central hospital. All enrolled patients were confirmed by tissue biopsies. Enzyme-linked immunosorbent assay (ELISA) was used to detect the 8-OHdG level in serum of all participants. Immunohistochemical SP method was used to measure the 8-OHdG level in tissue of all participants. Then we compared the serum and tissue 8-OHdG expression among all enrolled participants.Results: In patients with gastric carcinoma, tissue 8-OHdG expression was related to the differentiation of tumor tissue. 8-OHdG expression in high differentiated adenocarcinoma was significantly lower than that in low differentiated adenocarcinoma (p<0.05). There were no significant differences between 8-OHdG expression in patients with different age or gender (p<0.05).Conclusions: serum 8-OHdG expression was related to the development of gastric carcinoma, which showed that 8-OHdG can be a good serum biomarker for gastric carcinoma. Also, Tissue 8-OHdG expression was associated with the differentiation degree, clinical stage, invasion depth and lymphatic metastasis of gastric carcinoma. 8-OHdG may be used to predict severity of gastric carcinoma in tissue biopsy.
