目的:观察异硫氰酸苯乙酯(PEITC)对结肠癌细胞磷酸酯酶与张力蛋白同源物(PTEN)以及基质金属蛋白酶9(MMP-9)表达的影响。方法体外培养结肠癌细胞系SW480,分别用10、30、50μmol/L PEITC作用24 h,采用实时定量聚合酶链反应(real-time PCR)和荧光素酶报告基因分别检测PTEN 和MMP-9 mRNA 的表达;采用蛋白质印迹法(western blotting)检测MMP-9和PTEN蛋白表达。结果 PEITC可抑制MMP-9蛋白表达,同时能降低MMP-9的转录活性。此外,不同浓度PEITC处理后,可明显诱导细胞内PTEN的表达,其中50μmol/L PEITC处理后,PTEN mRNA含量可增高5.1倍。结论 PEITC能影响结肠癌细胞PTEN和MMP-9的表达。

목적:관찰이류청산분을지(PEITC)대결장암세포린산지매여장력단백동원물(PTEN)이급기질금속단백매9(MMP-9)표체적영향。방법체외배양결장암세포계SW480,분별용10、30、50μmol/L PEITC작용24 h,채용실시정량취합매련반응(real-time PCR)화형광소매보고기인분별검측PTEN 화MMP-9 mRNA 적표체;채용단백질인적법(western blotting)검측MMP-9화PTEN단백표체。결과 PEITC가억제MMP-9단백표체,동시능강저MMP-9적전록활성。차외,불동농도PEITC처리후,가명현유도세포내PTEN적표체,기중50μmol/L PEITC처리후,PTEN mRNA함량가증고5.1배。결론 PEITC능영향결장암세포PTEN화MMP-9적표체。
Objective To investigated the effect of phenethyl isothiocyanate (PEITC) on the expression of phosphatase and tensin homologue(PTEN) and matrix metalloproteinase-9(MMP-9) in colon cancer cells. Methods The colon cancer cell line SW480 was cultured in vitro,and treated with 10,30,50μmol/L PEITC for 24 h respectively. The mRNA expressions of MMP-9 and PTEN were detected by Real-time PCR and luciferase reporter gene respectively ,and the protein expressions of MMP-9 and PTEN protein were detected by Western blotting. Results PEITC could inhibit the protein expression of MMP-9 and reduce its transcriptional activity. In addition ,different concentrations of PEITC could markedly induced expression of PTEN in SW480 cells,and the mRNA contents of PTEN had 5.1-fold increase after treatment of 50μmol/L PEITC. Conclusion PEITC has influ-ence on the expression of MMP-9 and PTEN in colon cancer cells.
