目的:观察母鼠孕期和哺乳期暴露壬基酚(NP)对其雄性仔鼠脑皮质肿瘤坏死因子-α(TNF-α)表达的影响。方法根据妊娠时间将32只孕鼠随机分为对照组及NP低、中、高剂量暴露组,每组各8只;消化道暴露剂量分别为0、25、50、100 mg/(kg·d);暴露时间为受孕第6天到出生后21 d(仔鼠哺乳结束)。采用免疫组织化学方法检测仔鼠出生后21、60 d脑皮质TNF-α表达水平,分析其表达水平与NP暴露之间的关系。结果与对照组比较,NP低、中、高剂量暴露组仔鼠出生后21、60 d TNF-α表达水平增高。结论 NP孕期暴露使仔鼠脑皮质TNF-α表达水平增加,炎性反应可能是NP引起仔鼠脑皮质损伤的机制。

목적:관찰모서잉기화포유기폭로임기분(NP)대기웅성자서뇌피질종류배사인자-α(TNF-α)표체적영향。방법근거임신시간장32지잉서수궤분위대조조급NP저、중、고제량폭로조,매조각8지;소화도폭로제량분별위0、25、50、100 mg/(kg·d);폭로시간위수잉제6천도출생후21 d(자서포유결속)。채용면역조직화학방법검측자서출생후21、60 d뇌피질TNF-α표체수평,분석기표체수평여NP폭로지간적관계。결과여대조조비교,NP저、중、고제량폭로조자서출생후21、60 d TNF-α표체수평증고。결론 NP잉기폭로사자서뇌피질TNF-α표체수평증가,염성반응가능시NP인기자서뇌피질손상적궤제。
Objective To observe the influence of exposure to p-nonylphenol(NP) during pregnancy and lactation on tumor necrosis factor(TNF)-αin the cortex of male rat pups. Methods According to the pregnancy time,32 pregnant rats were randomly divided into the control group and the exposure group (of low,middle,high dose). Exposure dosages of digestive tract were 0,25,50,100 mg/(kg·d) respectively. Exposure duration was from 6th day of conception to the 21st day after birth (end of the lactation for newborn rats). Immunohistochemistry was adopted to determine the expression level of TNF-αin the cortex of male rats pups on the 21st day and 60th day after birth ,and the association between NP exposure and these alterations in TNF-αwas analyzed as well. Results Compared with the control group,the expression level of TNF-αin exposure group(of low,middle,high dose) increased on the 21st day and 60th day after birth. Conclusion Exposure to NP during gestation might induce an increase of TNF-αexpression levels in the cortex of pup rats,and the inflammatory reaction might damage mechanism of the cortex of pup rats.
