目的:探讨4-羟基-2,2,6,6-四甲基哌啶(Tempol)对肝硬化大鼠肝门阻断后肠道细菌移位和内毒素血症的作用。方法选择SD大鼠30只,采用四氯化碳诱导行肝硬化建模,将建模大鼠随机均分为假手术组(不阻断肝门)、对照组(阻断肝门)、实验组(阻断肝门前后静注Tempol )。3组均取门静脉、腔静脉血检测血清内毒素,取肠系膜淋巴结、肝组织、肺组织行细菌培养,以及小肠黏膜形态与细胞凋亡检测。结果与假手术组及对照组比较,实验组可明显减少肝门阻断后的肠道细菌移位及血清内毒素水平( P均<0.05)。结论 Tempo1可有效地预防肝硬化大鼠肝门阻断后的肠道细菌移位和内毒素血症。

목적:탐토4-간기-2,2,6,6-사갑기고정(Tempol)대간경화대서간문조단후장도세균이위화내독소혈증적작용。방법선택SD대서30지,채용사록화탄유도행간경화건모,장건모대서수궤균분위가수술조(불조단간문)、대조조(조단간문)、실험조(조단간문전후정주Tempol )。3조균취문정맥、강정맥혈검측혈청내독소,취장계막림파결、간조직、폐조직행세균배양,이급소장점막형태여세포조망검측。결과여가수술조급대조조비교,실험조가명현감소간문조단후적장도세균이위급혈청내독소수평( P균<0.05)。결론 Tempo1가유효지예방간경화대서간문조단후적장도세균이위화내독소혈증。
Objective To investigate the effect of 4-hydroxy-2.2.6.6-tetramethyl-piperidinyloxy ( Tempol ) on the bacterial translocation from the gut and the endotoxin after portal hepatic obstruction of liver cirrhosis in rats .Methods Selecting 30 SD rats and using the CCl 4 to induce liver cirrhosis , and then dividing them into the sham operation group ( not blocking the porta hepatis ) , control group ( blocking the porta hepatis ) and experimental group which was injected Tempol before and after blocking the porta hepatis .The serum endotoxin level from the portal vein and vena venous blood of the three group was detected , and we took the mesenteric lymph nodes , liver and lung tissue to conduct the bacterial culture as well as the detections of small intestinal mucosa morphology and apoptosis .Results Compared with the sham operation group and the control group , the intestinal bacterial translocation and the level of serum endotoxin were decreased in the ex -perimental group (all P<0.05).Conclusion Tempol has the preventive effect on the bacterial translocation and endotox-in after hepatic portal obstruction in cirrhotic rats .
