Bushen Rougan decoction%liver fibrosis in rats%α-SMA protein
目的:观察补肾柔肝方对肝纤维化大鼠肝组织病理学的影响。方法 SD大鼠随机分为空白对照组8只、模型组15只、秋水仙碱组15只、补肾柔肝方组15只。均以标准鼠食饲养,自由饮水,除空白对照组外,其余各组大鼠采用CCl4造模8周制备肝纤维化模型。从第9周开始,秋水仙碱组、补肾柔肝方组灌胃相应药物,空白对照组、模型组灌胃等体积生理盐水,均连续灌胃8周。末次给药后2 h,取肝组织进行HE、天狼猩红染色,评价肝细胞变性与胶原纤维增生程度。分别采用RT-PCR法和蛋白印迹法检测肝组织α-SMA表达情况。结果补肾柔肝方组大鼠肝脏的纤维化程度明显低于模型组;与空白对照组相比,模型组大鼠的α-SMA蛋白表达明显升高( P<0.01);与模型组相比,秋水仙碱组及补肾柔肝方组α-SMA蛋白表达水平有所降低(P均<0.05)。结论补肾柔肝方能够降低大鼠肝脏内α-SMA蛋白的表达水平,具有明确的抗肝纤维化作用。

목적:관찰보신유간방대간섬유화대서간조직병이학적영향。방법 SD대서수궤분위공백대조조8지、모형조15지、추수선감조15지、보신유간방조15지。균이표준서식사양,자유음수,제공백대조조외,기여각조대서채용CCl4조모8주제비간섬유화모형。종제9주개시,추수선감조、보신유간방조관위상응약물,공백대조조、모형조관위등체적생리염수,균련속관위8주。말차급약후2 h,취간조직진행HE、천랑성홍염색,평개간세포변성여효원섬유증생정도。분별채용RT-PCR법화단백인적법검측간조직α-SMA표체정황。결과보신유간방조대서간장적섬유화정도명현저우모형조;여공백대조조상비,모형조대서적α-SMA단백표체명현승고( P<0.01);여모형조상비,추수선감조급보신유간방조α-SMA단백표체수평유소강저(P균<0.05)。결론보신유간방능구강저대서간장내α-SMA단백적표체수평,구유명학적항간섬유화작용。
Objective It is to observe the effect of Bushen Rougan decoction on liver tissue pathology in rats with liver fi-brosis.Methods 53 SD rats were randomly divided into blank control group with 8 rats, model group with 15 rats, colchicina group with 15 rats and Bushen Rougan decoction group with 15 rats.All the rats were fed with standard rat food and given free drinking water.Except for the blank control group, the rats in the other groups were given CCl4 for 8 weeks to establish liver fibrosis models.The rats in colchicina group and Bushen Rougan group were given the respective drugs by intragastric adminis-tration on the ninth week, and the rats in blank group and model group were given the same volume of normal saline intragas-tric administration.In 2 h after the last intragastric administration, liver tissue of the rats were gotten to do liver tissue HE, sirius red staining to evaluate the degree of liver cell degeneration of collagen fiber hyperplasia, and the expression ofα-SMA in liver tissue was detected by PCR method and Westem blotting.Results The degree of fibrosis of the rats in Bushen Rougan decoction group was significantly lower than that of model group.Compared with the normal group, the expression ofα-SMA protein increased significantly in model group(P<0.01); compared with the rats in the model group, α-SMA protein ex-pression in colchicine group and Bushen Rougan decoction group decreased (P<0.05).Conclusion Bushen Rougan decoc-tion can reduce the expression level of α-SMA protein in rats with liver fibrosis and has definite effect of anti liver fibrosis.
