目的:探索葛根中有效成分葛根素的纯化方法,制备葛根提取物,提取物中葛根素含量应不得少于40%。方法通过对比试验确定树脂型号、用量及上样流速,用正交试验法确定树脂洗脱的条件。结果葛根经提取浓缩过滤后,上D-101大孔树脂吸附,1 g葛根药材(葛根素含量为2.71%)的提取液需用2.7 g D-101大孔树脂吸附,选择12 mL/min流速作为上样速度,选择20 mL/min水洗10倍柱体积的水清洗,以25%乙醇洗脱,收集洗脱液,浓缩至稠膏,减压干燥,所得葛根提取物葛根素含量均在40%以上。结论该法提取葛根中有效成分葛根素较稳定、可靠,适合工业生产。

목적:탐색갈근중유효성분갈근소적순화방법,제비갈근제취물,제취물중갈근소함량응불득소우40%。방법통과대비시험학정수지형호、용량급상양류속,용정교시험법학정수지세탈적조건。결과갈근경제취농축과려후,상D-101대공수지흡부,1 g갈근약재(갈근소함량위2.71%)적제취액수용2.7 g D-101대공수지흡부,선택12 mL/min류속작위상양속도,선택20 mL/min수세10배주체적적수청세,이25%을순세탈,수집세탈액,농축지주고,감압간조,소득갈근제취물갈근소함량균재40%이상。결론해법제취갈근중유효성분갈근소교은정、가고,괄합공업생산。
Objective To explore the purification method of effective components pueraria in radix puerariae.The puer-aria in the extraction of radix puerariae shall not be less than 40%. Methods It was determined the type and amount of resin , sample flow rate by contrast testing and the resin elution conditions were confirmed by orthogonal testing method. Results The ex traction and concentration of radix puerariae after filtering ,D-101 macroporous resin was used to absorb. The extraction (2.71%) of 1 g radix puerariae was prepared and adsorbed with 2.7 g D-101 macroporous resin by 12 mL/min.Washing by water with 20 mL/min and 10-time column volume,using 25%ethanol elution,collecting the eluted liquid,concentrating,vacuum dry-ing,the pueraria extract of the puerarin content should be more than 40%. Conclusion This process abstracting the effective comporent,namely pueraria technique are stable and reliable being suitable for industrial production.
