目的:研究人结肠癌细胞系SW﹣480、HT﹣29中肿瘤干细胞干性标记物CD44蛋白受阿司匹林不同药物浓度的影响变化,从而探讨阿司匹林是如何预防结直肠癌的发生。方法采用激光扫描共聚焦显微技术:免疫荧光双标染色法检测结肠癌细胞系SW﹣480、HT﹣29中CD44的表达;用Western blotting检测不同浓度阿司匹林(0、2.5、5.0、10.0 mmol/L,即分别为空白对照组、低浓度组、中浓度组、高浓度组)作用下,CD44在COX﹣2阴性表达的SW﹣480细胞及COX﹣2高表达的HT﹣29细胞表达影响。结果 SW﹣480、HT﹣29中存在CD44阳性细胞,2种细胞表达的CD44荧光值比较,差异有统计学意义(P<0.05);Western blotting结果显示阿司匹林能抑制结肠癌细胞系SW﹣480、HT﹣29中CD44的表达,且具有明显的量﹣效关系(P<0.05),2种细胞组间比较,差异无统计学意义(P>0.05)。结论结肠癌细胞系SW﹣480、HT﹣29中存在CD44阳性细胞,阿司匹林能抑制肿瘤干细胞干性标记物CD44的表达,其抑制可能是通过COX﹣2及非COX﹣2途径,该抑制作用可能是阿司匹林能预防结肠癌的原因之一。

목적:연구인결장암세포계SW﹣480、HT﹣29중종류간세포간성표기물CD44단백수아사필림불동약물농도적영향변화,종이탐토아사필림시여하예방결직장암적발생。방법채용격광소묘공취초현미기술:면역형광쌍표염색법검측결장암세포계SW﹣480、HT﹣29중CD44적표체;용Western blotting검측불동농도아사필림(0、2.5、5.0、10.0 mmol/L,즉분별위공백대조조、저농도조、중농도조、고농도조)작용하,CD44재COX﹣2음성표체적SW﹣480세포급COX﹣2고표체적HT﹣29세포표체영향。결과 SW﹣480、HT﹣29중존재CD44양성세포,2충세포표체적CD44형광치비교,차이유통계학의의(P<0.05);Western blotting결과현시아사필림능억제결장암세포계SW﹣480、HT﹣29중CD44적표체,차구유명현적량﹣효관계(P<0.05),2충세포조간비교,차이무통계학의의(P>0.05)。결론결장암세포계SW﹣480、HT﹣29중존재CD44양성세포,아사필림능억제종류간세포간성표기물CD44적표체,기억제가능시통과COX﹣2급비COX﹣2도경,해억제작용가능시아사필림능예방결장암적원인지일。
Objective To investigate the effects of aspirin affected by changes in the concentration of different drugs on CD44,which was supposed to be the marker of tumor stem cells in colon cancer cell line SW﹣480 and HT﹣29 so as to explore how to prevent the occurrence of colorectal cancer with aspirin. Methods The expression of CD44 in the colon cancer cell line SW﹣480 and HT﹣29 cells was observed by laser scanning confocal microscopy and immunofluorescence double staining. The expres﹣sion of CD44 protein in human colon cancer SW480(negative expression of COX﹣2) and HT﹣29(high expression of COX﹣2) cells with aspirin at different concentrations(0,2.5,5.0,10.0 mmol/L,namely the control group,the low﹣concentration group,the middle﹣concentration group and the high﹣concentration group) were detected by Western blotting. Results CD44+cells existed the SW﹣480 and HT﹣29 cells.There were statistical significance fluorescence value expressed by two kinds of cells. The difference had sta﹣tistical significance(P<0.05). Asprin could inhibit the expression of colon cancer cell line SW﹣480 and HT﹣29 by Western blotting, which showed an obvious dose﹣effect relationship (P<0.05). There were no statistical significance in difference between the two kinds of cells(P>0.05). Conclusion CD44+cells exist in the SW﹣480 and HT﹣29 cells. Aspirin may inhibit the expression of CD44 protein,the tumor stem maker in the colon cancer cell line SW﹣480 and HT﹣29,which was supposed to be acted by COX﹣2 and non COX﹣2 pathways. Asprin is probably one of factors to prevent colorectal cancer.
